| Objective:By researching the expression of the PTPRO Gene promoter methylation and the PTPRO gene m RNA in colorectal cancer tissue and normal tissue adjacent to carcinoma,to analysis differential expression of PTPRO in colorectal cancer tissue and normal tissue adjacent to carcinoma,and finding out the relationship between PTPRO and clinicopathologic characteristics of colorectal cancer to explore its role in colorectal cancer occurrence, development and its application in colorectal cancer diagnosis and prognosis.Methods:To collect 30 fresh specimens of colorectal cancer tissue which diagnosed by pathological and 30 normal colon tissue which are more than 5 cm distance tissue adjacent to carcinoma that from hunan province people’s hospital between April 2014 and November 2014 by surgical resection.Using Methylation specific PCR(MSP) and RT-PCR to detected PTPRO gene promoter methylation status and m RNA’s expression in 30 cases of colorectal cancer tissue and in normal tissue adjacent to carcinoma. analyzing the difference of PTPRO expressed in colorectal cancer tissue and normal tissue adjacent to carcinoma and its relationship with clinical pathology.Results:1.Colorectal cancer group of PTPRO m RNA expression level [0.2469(0.1118/0.3494)]are lower Normal e. group adjacent to carcinom PTPRO m RNA expression level [0.8998((0.4901/1.1253)], The difference is statistically significant(P < 0.05).2.Colorectal cancer tissues PTPRO methylation rate of 76.67%(23/30) was significantly higher in normal tissue adjacent to carcinoma(0/30).The difference was statistically significant(P < 0.05).3. The results of the relationship between PTPRO methylation positive expression rate and clinical pathologic features:The PTPRO methylation positive expression rate of Tumor size greater than or equal to 3 cm group(88%) are higher tumor size is smaller than 3 cm group(20%);Infiltration to size film and size film group(85.2%)are higher Into the submucosa below to serous membrane group(0%);TNM staging â…¢+â…£group(100%)are higher â… + â…¡ group(50%);Poorly differentiated group(100%) are higher high differentiation group(66.7%). With lymph node metastasis group(93.3%) are higher with no lymph node metastasis group(60%).The difference is statistically significant(P < 0.05).Conclusion:1.PTPRO gene promoter methylation are highly expressed in colorectal cancer, the expression of PTPRO gene m RNA in Methylation sample are obviously inhibited.,prompt PTPRO methylation is associated with the development of colorectal cancer.2.PTPRO gene methylation rate is connected with tumor size, TNM stages,differentiation degree, infiltration depth, with or without lymph node metastasis,prompt PTPRO methylation may be biological indicator to clinical diagnosis and prognosis. |
