Objective:To investigate the effect of BMSCs transplantation on adrenocorticotropic hormone( ACTH、CORT、NE and E) and AMPA receptors in rats with SCI and elucidate the potential anti-chronic stress mechanism of BMSCs transplantation in treating rats with spinal cord injury.Methods:48 adult male SD rats were equally divided into three groups:A control group,B model group and C treatment group.For Group B and C, lower thoracic SCI model was constructed(T10-11). For Group A, only laminectomy was performed. After 7 days, 100 u L Hank ’ s buffer suspension containing 106 BMSCs was injected into L4-5 intervertebral space of rats in group A and C. While for group B,only the equal volume of Hank’ s buffer was used. Meanwhile, BBB scale was also performed to evaluate preperative and postoperative hindlimb motor function of rats. The rats were sacrificed on 14 and 28 days after transplantation. The blood samples were collected for ELISA(ACTH,CORT,NE,E)and brains were harvested for immunohistochemical assay(Glu R1,2).Results:After BMSCs transplantation,the motor function of hindlimbs of rats in group C recovered gradually.14 d and 28 d after transplantation, the plasma ACTH content in group B and C was higher than that in group A,there was a significant difference between group B and A,group C and A(P<0.05).After transplantation of 28 d compared with 14 d, the serum CORT content was found to be elevated in group B and C and was higher than that in group A, there was a significant difference between group B and C(P<0.05),meanwhile,the serum CORT content in group C was lower than that in group B,there was a significant difference between group B and C(P<0.05).14 d and 28 d after transplantation, the plasma NE,E content in group B and C was higher than that in group A,and the NE,E content in group C was lower than that in group B,there was a significant difference between group B and C( P<0.05.14 d and 28 d after transplantation,the immunohistochemical assay suggested the number of cells expressing positively Glur1,2 receptor(from AMPA channel) in group C was lower than in group B,there were significant difference between 11 sets of data(P < 0.05); the count of positive cells in group B was higher than that in group A, there were significant difference between 6 sets of data(P < 0.05);the count of positive cells in group C was lower than that in group A,there were significant difference between 8 sets of data(P < 0.05);the other group did not see obvious change trend.Conclusion:BMSCs transplantation can improve the motor function of hind limbs in SCI rats. It can also contribute to the secretion of ACTH、CORT、NE and E. More importantly, it is able to regulate the central nervous AMPA channel Glu R1,Glu R2 receptor, suggesting its potential role in preventing chronic stress in SCI rats.
