Primary liver cancer is the malignant tumor originated in the liver, and it can cause a high mortality rate. Liver cancer, lung cancer and cancer of the stomach were called three major cancer in the world. Because the aetiological agent of liver cancer is more complex, and its specific pathogenesis is unclear. At present, the main treatment for liver cancer is surgery, transplantation, systemic chemotherapy, etc. But because of high costs, the lack of donor and postoperative recurrence etc, these factors serious impact on the treatment of liver cancer, choose better treatments appear imminent. Protein phosphorylation is one of the most important way of protein post-translational modifications, phosphorylation plays an important role in life activity process, which penetrate into the various activities of life, including cell proliferation, differentiation and development, cell apoptosis, signal conversion.The research phosphorylation proteomics of liver cancerous tissue cell can contributes to a greater understanding the protein phosphorylation effects on cells, find out the disease specific markers and targets, to further elucidate the molecular mechanism of the disease, provide a new strategy and provide a good theoretical basis for treatment of the disease.Objective:(1) Highly screen and identify differential protein expression and phosphorylated protein of patients’ cancerous tissue cells and cancer adjacent tissue cells. Drawing the phosphorylated protein profiling of cancerous tissue cells and cancer adjacent tissue cells to determine the sites of phosphorylation of these proteins. (2) According to the abundance distribution of phosphorylated protein, get the abundance analysis, and draw the phosphorylated protein abundance distribution map. (3) Protein and phosphorylation of protein function query expression differences and classified, the establishment of liver disease protein and phosphorylated protein group database from the whole.Method:Using protein enrichment technology iTRAQ protein markers and TiO2 phosphorylation in liquid phase, combined with tandem mass spectrometry analysis, to research the difference of eight liver cancer patients with liver cancer transplanted tissue and cancer adjacent tissue cells in protein and phosphorylation of protein expression, find out the differences between them.Results:The whole research find that the expression of protein and phosphoproteomic get a big Different between carcinoma tissue and cancer adjacent tissue.and the up-regulation expression of Protein in carcinoma tissue is about 158, dowm-regulation expression of Protein in carcinoma tissue is about 71.And total difference expression of phosphoproteomic is about 87, include 56 up-regulation expression and 31 dowm-regulation expression.Conclusion:the expression of Protein and Phosphoproteomic get a big Different between carcinoma tissue and cancer adjacent tissue.And with the ananlysis of the Gene Ontology (GO), we find that most of them are the part of the cell, and Involved in the biological regulation、 biological transformation、biological metabolism and some other biological processes.and they are maybe cause disease.
