The paper has studied the preparation and characterization of the water-soluble fluorescence polymer nanoparticles which was based on the carboxyl fluorescein and their application in cell fluorescence imaging and the single cell quantitative analysis in the first section.In this paper, the strong fluorescent polymers nanoparticles(FPNPs) were first prepared based on hydrophobic interaction of the as-synthesized strong fluorescent polymers with some extent hydrophobicity, and then PAAs were crosslinked on the surface of the FPNPs to prepare the stable and strong fluorescent nanoparticles(i.e. PAA surface-crosslinked FPNPs). The prepared nanoparticle had a special structure, and it consisted of the two parts of the fluorescent “rigid structure” and the “flexible structure”of the residual chains of PAAs. This kind of the special structure made the prepared nanoparticles have not only a smaller particle size and narrower particle distribution but also excellent stabilities for time, p H, plasma and anti-photobleaching. The residual chains of PAAs in the flexible structure not only easily labeled the prepared nanoparticles to the aptamers but also obviously decreased the steric hindrance of the labeled aptamers recognizing the huge-size target cells from large-size nanoparticles. Based on the aptamers labeled by the PAA surface-crosslinked FPNPs, this paper realized the high sensitivity of fluorescence imaging to human non-small lung cancer cell line A549, and the single cell quantitative analysis approach was also established with high detection sensitivity.The detective limit of 530 cells/m L for flow cytometry(R2=0.9914,S/N=3).The paper has studied the preparation and characterization of the water-soluble electrochemiluminescence polymer nanoparticles based on ruthenium in the second section.Firstly, The six coordination compound of rutheniumwith epoxy group Ru(bpy)2(phen-epoxide)(PF6)2(i.e.Ru-1)was synthesized which had the similar structure to three pyridine ruthenium. Then the water-soluble electrochemical luminescence polymer((Ru-1)n-PAA) was synthesized by the epoxy ring opening reaction between the epoxy group of the complexes and the carboxyl group of the PAA. Finally, The electrochemical luminescence polymer nanoparticles who had uniform distribution and 50 nm size was prepared by covalent cross-linking of the Ru-1-PAA under the reaction of the crosslinking agent XC- 105 by using the polymer molecule chain crosslinking method.
