Objective:To explore the impact of Pokemon on human breast cancer cell Proliferation and to analyze the correlationship of Pokemon with clinicopathologic features.Methods: the experiments includ two parts. The breast cancer cell MCF-7 in vitro culture was the research object of the first part, through constructing the lentivirus with the expression of Pokemon and then transfecting which into MCF-7 cells, the MCF-7 cell strain steadily expressing Pokemon was successfully established. Using MTT experiment method to detect the proliferation of MCF-7 cells and to draw the growth curve, At the same time using plate clone formation experiment to prove the effect of Pokemon on the proliferation of breast cancer cells, and then the relationship between overexpression of Pokemon and MCF-7 cell cycle distribution was detected by the flow cytometry. In the second section, through tissue chip and immunohistochemistry staining method to unveil the differential expression of Pokemon in 50 cases of breast carcinoma in situ tissues and lymph node tissues in corresponding, and to analyze its relationship with clinical pathology.Results : 1) The effect of Pokemon on the proliferation of MCF-7 breast cancer cells:①The expression of Pokemon decreased MCF-7 cell proliferous ability. ② Pokemon reduced the colony formation ability of MCF-7 cells. ③The MCF-7 cells were arrested in G1 phase of Pokemon. 2)The positive staining signals of Pokemon are subject to the nucleus colored,showing light brown, brown, brown granules. ①The expression of Pokemon in breast cancer is mainly in the nucleus, which including 4 cases of lymph node tissue specimens were accompanied by intracytoplasmic staining; There were 35 cases of positive expression of Pokemon in 50 cases of carcinoma in situtissues, the positive rate was 70%. There were 34 patients with Pokemon positive expression in the 50 cases of matched lymph node tissue samples, the positive rate was 68%.Pokemon in the two groups both presented high expression,But the difference between them was not statistically significant(p> 0.05). ②In situ breast cancer tissues, there are some correlations between the expression of Pokemon and the expression of ER or PR.The expression of Pokemon was positively correlated with increased expression of ER, It’s also the same relationship with PR,the difference was statistically significant between them(p<0.05),but it has a greater relevance with ER(R=0.515). In Lymph nodes of breast cancer tissue, the expression of Pokemon was positively correlated with gradually increased ER, the difference was statistically significant(p<0.05), But it did not relate to PR(p> 0.05). ③In situ breast cancer tissue: Age of onset:in the following groups under 50 years old and over 50 have positive expression rate of Pokemon,Respectively were 72.7% and 67.9%,between the two was no significant difference(p> 0.05). Nuclear grade:In cancer tissues,The cell nuclear grade of differentiation is divided into 1 scale, 2 scale, 3 scale,then Pokemon positive expression rate were respectively 83.3%, 66.7%, 50%, compare the differences amongthe three groups were not statistically significant(p> 0.05). ④Lymph nodes in breast cancer tissue : Age : in the following groups under 50 years old and over 50 have positive expression rate of Pokemon,Respectively were 53.6% and 78.6%,between the two was no significant difference(p> 0.05). Nuclear grade:In cancer tissues,The cell nuclear grade of differentiation is divided into 1 scale, 2 scale, 3 scale,then Pokemon positive expression rate were respectively 83.3%, 72.2%, 25%, compare the differences amongthe three groups were not statistically significant(p> 0.05).Conclusion:1)Pokemon inhibited the proliferation of MCF-7 cells, its mechanism might be MCF-7 cells was blocked in G1 phase by Pokemon. cloud block MCF-7 cells at G1 phase.2)There is a positive correlation between the expression of Pokemon and the increasing of ER in situ breast carcinoma,it’s similar to PR. In lymph nodes of breast cancer tissue, there is a positive correlation between the expression of Pokemon and the increasing of ER.
