Objective To observe the inhibitory effect and mechanism of Curcumin in combination with Imatinib on proliferation and apoptosis in chordoma cells.Methods The chordoma cell line CM-319 was cultured in vitro and treated by different concentration of Curcumin and(or) Imatinib for 24~72h. Proliferation of cells were detected by WST-1 methods, expression of m RNA of platelet-derived growth factor receptor-β(PDGFR-β) was detected by real-time PCR; Phosphorylation of Akt and expression of protein of caspase-3,bcl-2 and PDGFR-β were measured by Western blot. Apoptosis of cells was detected by flow cytometry.Results 1、As the concentration of Curcumin and(or) Imatinib increased, the inhibitory effect on CM-319 proliferation was also gradually strengthened. Then the survival rate reached to 50%, the combination index was 0.86. 2、10μmol/L of Curcumin could not decrease the expression of PDGFR-β m RNA of CM-319 cells, but could inhibit the expression level of PDGFR-β protein. As the increase of curcumin concentration, the expression level of m RNA and PDGFR-β was gradually decreased. The expression of PDGFR-β m RNA and protein was not affected by treatment of 1μmol/L of Imatinib, but it could significantly decrease PDGFR-β m RNA and the expression level of protein after used in combination with Curcumin for 24 h. 3、Co-incubation of 1μmol/L Imatinib and 10μmol/L Curcumin could decrease the level of the phosphorylation of Akt. 4、1μmol/L of Imatinib and 10μmol/L of Curcumin had no effect on CM-319 apoptosis, the number of CM-319 apoptosis only slightly increase after deal with 20μmol/L Curcumin, when the Curcumin was used in combination with Imatinib, the apoptosis of cells proportion was significantly increased.Meanwhile, level of caspase-3 was rised obviously,but level of bcl-2 was decreased.Conclusion 1. It is probable that Imatinib and Curcumin can inhibit chordoma cells proliferation by inhibition of the expression of PDGF-β, phosphorylation of PI3K/Akt. 2. Combination of Imatinib and Curcumin can induce chordoma cells apoptosis induced by down-regulation of bcl-2 and up-regulation of caspase-3.
