Objective 1. The research about the effect of the Xi Huang Wan to the epidermis staphylococcus biofilm in vitro; 2. Xi Huang Wan will be take application to the model of the artificial knee joint of the rabbit which the prosthesis infect by the epidermis staphylococcus, research Xi Huang Wan influen Xi Huang Wan on rabbit knee prosthesis in epidermis staphylococcus infection after the influence of bacterial biofilmce on epidermis staphylococcus bacteria biofilm in vivo.Methods 1. The research about the Xi Huang Wan to the effective of the epidermis staphylococcus bacteria biofilm in vitro. Through the tset the ability of the epidermis staphylococcus form ATCC35984 bacterial biofilm, observe Xi Huang Wan on the impact of epidermis staphylococcus ESS; By using the methods about the fluorescent microscope and laser confocal microscope to observe the shape of BF, research Xi Huang Wan in the effective of the epidermis staphylococcus bacteria biofilm in vitro. 2, The research about the Xi Huang Wan to the effective of the epidermis staphylococcus bacteria biofilm in vivo. Use the New Zealand white rabbit to build a knee prosthesis model which infect by the epidermis staphylococcus bacteria biofilm all around about it, inject the epidermis staphylococcus ATCC35984 to the articular cavity of the rabbit after one week, then the model which we need will be found. One week after inoculation, confirmed infection with building successful, the rabbit can be divided into four groups at random, control group, the Xi Huang Wan treatment group and levofloxacin in treatment group and combination group, though feeding, observing animals systemic condition by inspect the content of the WBC, CRP, ESR、count the bacterial biofilm that stick to the surface of prosthesis、observe the shape of BF by the CLSM、bacterial culture or pathological section in the articular cavity and etc,to observed Xi Huang Wan in the effective of knee prosthesis which all around inject by the epidermis staphylococcus bacteria biofilm.Results 1. In vitro experiments, it showed that epidermis staphylococcus ATCC35984 have strong ability to form the BF; Xi Huang Wan can reduce the ESS which secrete by epidermis staphylococcus ATCC35984 bacterial biofilm, Xi Huang Wan can enhance levofloxacin antimicrobial effects that under the epidermis staphylococcus bacteria biofilm. 2. After intervention therapy, Xi Huang Wan group and levofloxacin group and combined group of animals BF are reduced, the joint group of the most obvious. In vivo tests it is concluded that the Xi Huang Wan combined levofloxacin is effective prevention and treatment of joint prosthesis epidermis staphylococcus infection, Xi Huang Wan can enhance levofloxacin antiseptic effect.Conclusion 1. In vitro environment test show that the Xi Huang Wan of epidermis staphylococcus bacteria biofilm inhibition 2. In vivo environment tests showed that not only the epidermis staphylococcus form bacterial biofilm can be restrain by Xi Huang Wan, but also it combine with levofloxacin can enhance the effect of anti-infection.
