Diagnostics of clinical laboratory Postgraduate: Wangzuolong Supervisor: Ma Li [Abstract] Objective: Discusses the relationship of Th9 cells、CD4+CD134 cells、HLA-B27(human leukocyte antigen B27)fluorescence intensity with the on come and development of Ankylosing Spondylitis,By the detection of T helper cells 9(Th9)、 CD4+CD134 cells、HLA-B27 fluorescence intensity in the peripheral blood of Ankylosing Spondylitis(AS)patients. Methods:Application of Flow Cytometry(FCM) and Intracellular Cytokine Staining(ICS)test in the experiment AS group and healthy controls Th9 cells、CD4+CD134 cells、HLA-B27 fluorescence intensity in peripheral blood,And analyze them with the AS inflammatory activity correlation.Results:1.Th9 cells percentage of the Peripheral blood in the AS disease group, low activity and high activity group was 4.37%, 2.96% and 5.8%, respectively, were higher than healthy controls(0.44%),The difference was statistically significant(P <0.01);AS low activity and high activity group comparison, AS low active group is lower than the high active group, the difference was statistically significant(P <0.01);The CD4+CD134+ cell percentage in Peripheral blood in the AS disease group, low activity and high activity group were 5.54%, 3.74%, and 7.33% were increased, were higher than healthy controls(0.9%), the difference was statistically significant(P<0.01);AS low activity and high activity group comparison, AS low active group is lower than the high active group, the difference was statistically significant difference(P < 0.01).HLA-B27 fluorescence intensity of the Peripheral blood in the AS disease group, low activity and high activity group was 163.1, 161.4 and 164.7, respectively, were higher than healthy controls 84.5,The difference was statistically significant(P < 0.01);AS low activity and high activity group comparison, AS low active group is lower than the high active group, the difference was not statistically significant(P > 0.01).2.The AS patients Th9 cells、CD4+CD134+cells with AS active correlation analysis showed that Th9 cells、CD4+CD134 cells percentage and activity(BASDI) were positively correlated(r=0.897,0.969,P<0.01);The AS patients HLA-B27 fluorescence intensity with AS active correlation analysis showed that HLA-B27 fluorescenceintensity activity(BASDI) were no correlated(r=0,P> 0.01). Conclusion: 1.The Th9 and CD4+CD134+ cells in the peripheral blood of AS patients involved in the oncome and development of the AS disease.2、Detection of Th9、CD4+CD134+ cells in peripheral blood contribute to the AS patients judgment, is expected to become the AS progression monitoring measurements.3. HLA-B27 fluorescence intensity has a guiding significance for the diagnosis of the AS,HLA-B27 fluorescence intensity activity were no correlated.
