ObjectiveTo observe effects of Sanao Decoction on TGF-β1 and NGF during airway remodeling in asthmatic mice model, and explore the underlying intervention mechanism on the process of airway remodeling. Finally, the data can provide references for expanding clinical applications and carrying out basic experiments of Sanao Decoction.MethodsForty SD rats were averagely divided into four groups, and they were normal control group, asthma model group, budesonide group(2.5x10-4g/kg) and Sanao Decoction group(39g/kg). The asthma model was established with the method of OVA sensitization united with challenge. Finally, the count of EOS in peripheral blood was detected; the thickness of airway wall and airway smooth muscle was measured; concentrations of TGF-β1 in BALF and serum were detected and expression levels of NGF in the lung tissue were detected with immunohistochemistry.Results1. Compared with normal control group, the thickness of airway wall and airway smooth muscle increased, the count of EOS in peripheral blood increased, concentrations of TGF-β1 in BALF and serum increased, expression levels of NGF in the lung tissue increased in asthma model group, and all the differences among them have statistical significance.2. Compared with asthma model group, the thickness of airway wall and airway smooth muscle decreased, the count of EOS in peripheral blood decreased, concentrations of TGF-β1 in BALF and serum decreased, expression levels of NGF in the lung tissue decreased both in budesonide group and Sanao Decoction group, and all the differences among them have statistical significance.3. Compared with budesonide group, the count of EOS in peripheral blood increased in Sanao Decoction group, and the difference had statistical significance; there existed no statistical significance among the thickness of airway wall and airway smooth muscle, concentrations of TGF-β1 in BALF and serum and expression levels of NGF in the lung tissue.Conclusions1. It indicates that the asthmatic mice model was successfully established with the method of OVA sensitization united with challenge.2. Sanao Decoction can inhibit the process of airway remodeling in asthmatic mice model effectively. And the mechanism involves three aspects. It refers to reducing the count of EOS, decreasing concentrations of TGF-β1 and down-regulating expression levels of NGF.3. The effects of Sanao Decoction on inhibiting the process of airway remodeling during asthma is similar to budesonide, and Sanao Decoction has a certain degree of advantages.
