Objective To explore the effects and underlying mechanisms of glucocorticoid(GC) in apoptosis of retinal ganglion cell(RGC) in vitro.Methods RGC were cultured in 4 groups for 3 days in vitro: control group, GC group(0.1 μmol/L cortisone), GC-si Ng R group(0.1 μmol/L cortisone + Ng R antisense nucleotide), GC-sc RNA group(0.1 μmol/L cortisone + scrambled nucleotide). The cell viability was detected by using methyl thiazolyl tetrazolium(MTT) methods, the morphological alterations were detected with inverted microscope, the apoptosis of RGC was observed by using hoechst 33342 staining, and the expression of Nogo receptor(Ng R) was revealed by Western blot.Results Control, GC, GC-sc RNA and GC-si Ng R groups presented cell viability with(100.0±0.0)%,(76.3±6.8)%,(79.4±9.0)% and(96.7±9.8)%, and showed expression of Ng R with(20.0±2.7)%,(736.3±1.8)%,(739.5±4.0)% and(718.7±2.5)%. Decreased cell viability and increased Ng R expression were detected in GC group and GC-sc RNA group(P<0.01, respectively), together with atrophic cell body. While, these alterations were not observed in GC-si Ng R group(P>0.05). RGC showed slightly blue by hoechst 33342 staining in control group and GC-si Ng R group, exhibiting bright blued apoptotic RGC in GC group and GC-sc RNA group.Conclusions Up-regulation of Ng R contributes, least in part, to glucocorticoids-induced apoptosis...
