Objective: The use of proteomics experiments, by liquid chromatography- mass spectrometry, constructs different protein in brain tissue of rats with acupuncture at back Shu Points insomnia model spectrum front, detect acupuncture improve sleep quality associated with which protein, effect changes generated prove pathological changes and body acupunct ure insomnia after, provide strong experimental evidence for acupuncture treatment insomnia Methods: Selected 150 SD rats of clean grade, male, weight 200 + 10 g,which were randomly divided into five groups.The observation after 7-day feeding showed that each rat was in good condition.Then each group was treated in the following method: blank control group did not do any related processing; normal bundle group accepted grabbing once a day,; the n ormal acupuncture group was treated with grabbing,and begun to accept acupuncture from t he third day,7 consecutive days in total; model application group was appied with PCPA inje cted intraperitoneally at the beginning stages, according to 1mL of injection for every 100 g,injection for 2 days, grabbing lasting 7 days and begining from the 3rd day,no acupuncture,lasting 9 dys in total;acupuncture treatment group was applied with PCPA by intraperitone al injection,according to 1mL of injection for every 100 g, injection for 2 days, and was appli ed with grabbing and acupuncture from the 3rd day,,acupuncture for 7days.The above after the experiment materials: selection of chloral hydrate anesthetized rats by intraperitoneal injection, the rats were decapitated after treatment, the rats head is placed in the Bingpan, rapid pry craniotomy with bone gently stripping out brain tissue by using shear operation; liquid chromatography- mass spectrometry technology was different between groups differences in protein expression analysis.Result: 1. the model control group and acupuncture treatment group rats model of suspension 24-36 h para chlorophenylalanine on first after intraperitoneal injection, the emergence of circadian activity rhythm disorder, daytime activity is frequent, some rats obvious emotional restlessness, through the contrast observation, and blank control group with significant difference. The model rats after acupuncture treatment group and acupuncture treatment, the amount of sleep and eating water quantity rules are slowly recovering. The normal bundle control subjects with normal acupuncture group of two rats showed no significant change in the law of life, sleep habits and diet.2. Proteins were identified to the experimental group for 2 to 3383(ProteinGroup), protein group number where each channel analysis of two biology have quantitative information is respectively 2743 and 2952. Five groups of samples in each channel, will mark 113, 114, 115, 116, 117 and the channel mean REF ratio, and analyzed the differences between the two groups of protein quantity is: between the A group and the D group measured between 163 differential protein, A group and E group measured between 203 differential protein, B group and E group were measured between 245 differential protein, D group and E group measured 118 differential protein(including number of differences between Ratio>+/-1.2 and protein expression by P value<0.05 screening).Conclusion: 1. experimental data shows, acupuncture at back Shu Points to prevent and cure the insomnia; acupuncture treatment of insomnia rats model, specific expression of the rat brain cathepsin generated solutions may be related with the mechanism of acupuncture in treating insomnia; there are certain differences in expression and using different stimulation methods produce protein.2.the model control group and acupuncture treatment group mass spectrometric analysis model was found after repeated some, a small part of the protein is relatively obvious differential expression, prove that differential expression of acupuncture can change the insomnia rat models of brain protein; for the differences detected protein, may be due to the generation after the acupuncture treatment of insomnia.
