Caffeine which becomes the most edible spirit of active substances widely exists in drinks and conventional drugs. In normal doses, it is good for movement and neural function. Long-term consumption of produce makes mild addiction. Once the outage of produce, people perform short a few days’headache or discomfort. Poisoning dose can cause clonic convulsion. Domestic researches on caffeine are more concentrated in soft drinks, tea, tablets and other carrier, but the research of caffeine and its metabolites in the hair is few. Since hair analysis can provide long-time personal use of the drug information, caffeine and its metabolites in the hair analysis for individual whether takes caffeine is of great significance.External caffeine can be adsorbed on the hair in various ways, it is easy to cause false positive results. Mistakenly identified as positive results can be divided into technical error checking and error explanation. Using some advanced MS detection can obtain higher accuracy and higher sensitivity, such as with quadrupole Mass Spectrometers or MS/MS ions well device used in positive or negative chemical ionization together, which reduce false positive error checking.Experiments with 0.1% sodium dodecyl sulfate (SDS),0.1% detergent, distilled water, acetone in turn remove caffeine which adhere on the surface of the hair. This experiment used trace chloroform solvent extraction of caffeine, theobromine and theophylline, which makes sample preparation is simple, timesaving and promotes the development of instrument analysis automation, can reduce the damage to operators. GC/MS with gas chromatography efficient separation and mass spectrometry of the characteristics of high sensitivity and high specificity can improve the efficiency of separation and analysis, and can check out the trace amounts of samples. The linear correlation coefficient of caffeine, theobromine and theophylline is more than 0.99.At the concentration of 100 μg/mL, the relative standard deviation of caffeine, theobromine and theophylline are 3.96%,3.21% and 3.36% respectively. At the concentration of 60 μg/mL, the relative recovery rate of caffeine, theobromine and theophylline was 95.70%,89.84% and 77.72%, respectively.
