[Objective] The research aims at summarizing tutor Professor Yang Minghui’s academic experience as well as clinical experience in treatment of Parkinson’s disease (PD). Through the establishment of an animal model of Parkinson’s disease, this research adopts the immunohistochemical method to observe changes in associated protein expression of rat’s brain tissue, and to explore the mechanism of tonifying kidney and invigorating blood circulation for PD treatment, which would provide an objective basis for the traditional Chinese medicine (TCM) treatment of PD.[Method] According to lectures of the tutor, clinical diagnosis, medical records and related documents, the research summarizes Professor’s academic experience particularly in the treatment of PD. The animal experiment has been done as follows:60 male SD rats were randomly divided into 10 as normal control group and 50 as model group. Rats in model group were enforced by means of 6-OHDA damaged method for the purpose of manufacturing PD model. Then 23 successful models were randomly divided into 10 as compared group and 13 as treatment group. During the period of 8 weeks, rats in treatment group had been continuously intragastrically administrated with Bushen Huoxue Granule(BSHXG); meanwhile, rats in the compared group had been treated with physiological saline. As well as protein expression of Fas, Fas-aasociated death domain (FADD), Bcl-2 and Bax in rat’s nigrostriatal system by the technique of immunohistochemical staining.[Results] Professor innovatively proposed that PD’s main pathogenesis is the deficiency of kidney and stagnation of blood. Years of application for the PD treatment of tonifying kidney and invigorating blood circulation has achieved significant clinical efficacy. Animal experiments confirmed that BSHXG can significantly improve the rotational behavior of PD rats. Immunohistochemistry results showed that in the model group, the positive cells expression rate (PI) of Fas in the substantia nigra and the striatum are 1.4±0.07,1.82±0.091 respectively, which are higher than normal control group (both P<0.01) and treatment group (both P<0.01). PI values of FADD in the substantia nigra and the striatum in the model group are 1.16±0.058,0.78±0.039 respectively, which are also higher than normal group (both P<0.01) and treatment group (both P<0.01). Meanwhile, Bcl-2 PI values in the substantia nigra and the striatum in the model group are 0.43±0.022,1.14±0.057, which are lower than the normal group (both P<0.01) and treatment group (both P<0.01); where as Bax PI values in the substantia nigra and the striatum are 2.07±0.104,2.86±0.143, higher than the normal group (both P<0.01) and treatment group (both P<0.01)[Conclusions] The treatment of tonifying kidney and invigorating blood circulation for PD syndrome applied by the tutor is clear in dialectic thinking, complete in treatment system, and confirmed in clinical efficacy. Experimental results show that the BSHXG can significantly improve the rotational behavior of PD rats, one of the possible mechanisms of BSHXG for PD treatment lies in lower the protein expression of Fas, FADD and Bax in rat’s nigrostriatal system, whereas increase Bcl-2 protein expression.
