Objective: To investigate the apoptosis effect of Protocadherin10 gene(PCDH10) on endometrial glandular cells and its mechanisms. Methods: The human endometrioid endometrial adenocarcinoma cell line HEC-1-B, KLE and AN3 CA was stably transfected with pcDNA3.1(+)PCDH10 or pcDNA3.1(+)vector by lipofectamine. For the following experiments, pcDNA3.1(+)vector plasmid transfected cells were used as control.The expressions of PCDH10 mRNA in the PCDH10-tansfected cells were detected by RT-PCR and real-time PCR. Cell proliferation after transfection was detected by MTT method.The cell apoptotic rate and cell cycle was assayed by flow cytometry. Results: After transfection with PCDH10 gene, it was showed that over-expression of PCDH10 gene resulted in obviously increased expressions of PCDH10 gene. MTT results showed PCDH10-AN3 CA and PCDH10-HEC-1-B cells proliferation was significantly1 suppressed. The flow cytometry assay revealed increased apoptosis of AN3 CA and HEC-1-B cells line transfected with PCDH10 gene(P<0.05) amd blocked of G1 phase of HEC-1-B and AN3 CA cells. Conclusion: 1. The PCDH10 gene plays a supressed role in EEC development. 2.The PCDH10 gene plays a promoting role in apoptosis and blotting of G1 phase of EEC cell line.
