objective:Combining the epidemiology and molecular methods to explore the epidemiology and subtypes of HIV-1 in Qinzhou City.Method:collecting the demography and epidemiology informatio n of 50 HIV-1 infected patients by using questionaire,extracting tota l virus RNA From each 5 to 10 ml Blood sample of each infected people.Amplifying the gas fragment by using nested reverse transcrip tase-polymerase chain reaction and identifying the PCR product, gen e sequencing was performed by microbial institute of epidemic of Beijing’s military academy,using Bio Edit、MEGA5.03 software、 Dis tance procedure 、MEGA5.03 software、Genotyping online to analyz e(Cutting, calibration,alignment,build system evolutionary tree and su btype analysis) the gene sequence.Results:1.a total of 50 blood samples were collected,the demography and epidemiology information are as follows(1)the Living District of 50 infected people including Qinnan zone of Qinzhou city, Qinbei zone and its Pubei 、linshan county.(2).the male/female of infected was 1.38:1,31 of 50 people were less than 40 years old,48 of these were from Han and Zhuang nationality.(3) sexual transmission and intravenous drug use accounted for 96% of all propagation paths.2.finally 36 strips of gene sequence were acquired after 50 blood samples were extracted total RNA,amplification by using nested reverse transcriptase-polymerase chain reaction. the amplification ratio was 72%.3.there recombinant virus including 01AE、07BC、08BC 3 we re identified by analyzing 36 gene sequence using NCBI online clas sification and certification with MEGA5.03 software Build system tr ee, in these 36 gene sequence, 01AE subtype counted 32 case(88.90%),07BC subtype counted 2 case(5.55%),08BC subtype counted2 case(5.55%),subtype genetic distance were 8.62%,6.47% and 5.73%.4.phyletic evolution analysis: 32 case 01AE subtype recombina nt strain and 01AE.TH.91.TN240.L11761 from Thai reference strai n held together, the discrete rate between them is minimal(9.12±1.76)%, 2 case 07BC subtype recombinant strain and 07BC.CN.97.97.CN001C54.AF286226 from Chinese reference strain held togethe r, the discrete rate between them is minimal(7.31±2.43)%.2case 08BC subtype recombinant strain and 08BC.CN.97.97.CNGX9F.A Y008717 from Chinese Guangxi BC recombinant strain held togethe r, the discrete rate between them is minimal(6.64±2.58)%.5.the distribution mode of transmission of different subtypes: 30 out of 32(2 out of 32) case of 01AE subtype infected had unprotected sex, and(intravenous drug),2 case of 07BC and 2 case of 08BC all had unprotected sex.Conclusion: 1.HIV-1 epidemic is comprehensive popular in all counties, male/female ratio tends to be balanced, the infected are mainly young adults less than 40 years old from Han and Zhuang nationality population. the main transmission routes are unprotected sex and intravenous drug.2.there are there prevalent subtypes(01AE、07BC、08BC)in infected people from Qinzhou City, especially 01AE subtype, and the 01AE subtype is spreading from high risk group to general population, which may be the main subtype of prevalent strain in Qinzhou City or Guangxi province.3.the period of each epidemic strain are speculated by gene disp ersion ratio analysis as follows:01AE 9-15 years,07BC 8-12 years,08BC 6-10 years.4.we revealed the diversity of HIV-1 prevalent strain from the perspective of molecular biology, the prevention and control of HIV-1 in Qinzhou City should be highly valued.
