| Candida albicans, a kind of opportunistic pathogenic fungi, is a part of the normal flora of the mucous membranes of the respiratory, gastrointestinal, and female genital tracts, and is almost universal in low numbers on healthy humans without causing harmful effects. However, when mucosal barriers are disrupted or the immune system is compromised Candida albicans could invade and cause disease.According to the survey, fungal infection has become one of the important causes of death in clinical patients, candidiasis is given priority to with fungal infection. Candida albicans infectionis the mainly pathogens of candida infection.Blood culture, the gold standard of traditional method of candida infection detection, has the obvious shortcomings such as the long cycle, which often delays the best time to treatment. By contrast, serological detection with the advantage of simple, effciency and accuracy is developed rapidly. As an immunodominant antigens, fructose-bisphosphate aldolase1(Fba1) draws more and more attentions. The body could produce the antibodies of Fba1 at the beginning of the candidiasis, the Fba1 protein could be used as an effective target for detecting of candidiasis.Phage could be displayed a single protein epitope on the phage surface, and use to serological detection for increasing the specificity, The combination of the phage display technology with recombinant protein technology can guarantee the detection rate and avoid the occurrence of a false positive result in the detection of candidiasis.In this study, a prokaryotic expression vector carrying the fructose-bisphosphate aldolase1 gene was constructed as well as a recombinant phage displayed its single epitope, the specificity of which was validated prior to be used in serum detection. The serum of healthy person and cancer patients were examined with the method of joint detection, and the results showed that the joint detection increased the sensitivity and specificity of the detection of candidiasis.An effective way to reduce the mortality of candidiasis is early diagnosis and early treatment. This study used serological detection method of fungi infection, which combines the recombinant protein technology with phage display technology, laid the foundation for the early diagnosis of candidiasis. |
