Objectives: To elucidate the role of phosphatidylinositol binding clathrin assembly protein(PICALM)gene rs541458 and rs3851179 as a potential cause of Alzheimer’s disease(AD) susceptibility in different ethnic groups. Methods: The study was performed uninterruptedly to choose 30 different ethnic AD patients and age-matched healthy controls in Dali University Hospital and Dali Psychiatric Hospital since October, 2011 to October, 2013. We diagnosed AD based on patients’ history, clinical symptoms, and a variety of psychiatric scales(CCMD-3, MMSE, ADL, HDS-R, CDR, HIS and HRSD). We were also collected the patient’s smoking history, drinking history, education level, fasting plasma glucose levels, fasting plasma lipid levels, vasculopathy, family history and other related information. By using salting out method, DNA was extracted from whole blood, and PICALM gene polymorphism rs541458 and rs3851179 were genotyped by using direct sequencing. We used chi-square test, analysis of covariance(ANCOVA) and Logistic regression analysis to discuss the relationship of rs541458 and rs3851179 PICALM gene polymorphism with Alzheimer’s disease. Results: 1、The genotype frequencies of CC, CT and TT with PICALM gene rs541458 polymorphism in the AD group were 30%, 46.7%, but 23.3%,23.3%, 53.4%, 23.3% in the control group. The genotype frequencies of CC, CT and TT with PICALM gene rs3851179 polymorphism in the AD group were 3.3%, 56.7%, 40%, but 6.7%, 56.7%, 36.7% in the control group. 2、PICALM gene rs541458 polymorphism genotypes and allele frequency distribution in different nationalities(Han, Yi, Bai and other nationalities) differences between AD patients and healthy controls had no statistical significance(P >0.05). PICALM rs3851179 polymorphism genotypes and allele frequency distribution in different nationalities(Han, Yi, Bai, other nationalities) differences between AD patients and healthy controls had no statistical significance(P >0.05). 3、Group AD patients with different genotypes and the spirit of the scale difference between the examination score were not statistically significant(P >0.05). 4、There was statistical significance between different ethnic groups in AD patients and control group of plasma lipid, smoking history, alcohol history, family history of differences(P <0.05). Conclusions: 1、Patients with AD in different nationalities(Han, Yi, Bai and other nationalities) of Dali compared with healthy controls, plasma glucose levels, plasma lipid levels are relatively high and have a higher proportion of drinking alcohol history and family history of dementia and have a lower proportion of smoking history. 2.The PICALM gene is not associated with Alzheimer’s disease in different nationalities(Han, Yi, Bai and other nationalities), so, we couldn’t consider that PICALM gene rs541458 and rs3851179 are risk factors for Alzheimer’s disease of different nationalities in Dali region.
