Objective: To investigate the inhibitory effects of Ginsenoside Rd(GS-Rd)on the apoptosis of U251 cells in vitro. Methods: The proliferation activity was detected by MTT assay. The group include blank control group, solvent group, GS-Rd low group(GS-Rd 20μmol/L), GS-Rd middle group(GS-Rd 40μmol/L), GS-Rd high group(GS-Rd 80μmol/L); ①The cell apoptosis of U251 were detected by flow cytometry.②The change in the cell growth and form of U251 were detected by fluorescence phase contrast microscope;③Eliasa assay was used to measure the telomerase activity in different group on 24 h, 48 h, 72 h.;④Real-time quantitative PCR was used to measure the expression of Bcl-2, h TERT and caspase-3 in different group on 48 h. Results: ①The proliferation of U251 cells was inhibited by Ginsenoside Rd with the concentration of 20 μmol/L to 80 μmol/L in dose dependent manner, based on IC50, 77.38μmol/L, the GS-Rd groups includ GS-Rd low group(GS-Rd 20μmol/L), GS-Rd middle group(GS-Rd 40μmol/L), GS-Rd high group(GS-Rd 80μmol/L)②GS-Rd with the concentration of 20 ~ 80μmol/L range can be induced U251 cell apoptosis rate,Compared with blank control group, the group of solvent could not enhance the expression of cell apoptosis rate(P > 0.05).Compared with blank control group, the group of GS-Rd 20μmol/L could enhance the expression of cell apoptosis rate(P < 0.05),Compared with blank control group, the group of GS-Rd 80μmol/L could significantly enhance the expression ofcell apoptosis rate(P < 0.01),③All dose groups of Ginsenoside Rd could induce apoptosis of U251 cell, cell form showed that the increase of Ginsenoside Rd concentration, the quantity of cells was decreased, and the cell appeared the obvious changs of apoptosis characeristics.③Compared with blank control group, the group of solvent on 12 h, 24 h and 48 h, could not enhance the expression of teleomerase(P > 0.05).④Compared with blank control group, the group of GS-Rd 20μmol/L on 24 h, 48 h could significantly enhance the expression ofteleomerase(P < 0.01),but on 12 h could not enhance the expression of teleomerase(P > 0.05)⑤Compared with blank control group, the group of GS-Rd 40μmol/L and 80μmol/L on 12 h, 24 h, 48 hcould enhance the expression of teleomerase; ⑥Compared with blank control group, all dose groups of GS-Rd could increase the gene expression of caspase-3, significantly enhance the gene expression of caspase-3 m RNA( P < 0.01); decrease the gene expression of Bcl-2 m RNA and h TERT m RNA, significantly enhance the gene expression of Bcl-2 m RNA and h TERT m RNA( P < 0.01).Conclusion: The Ginsenoside Rd can remarkably restrain the multiplication of U251 glioma and promote the cell to apoptosis, and antitumor mechanisms could related to restrained its telomerase activity, down-regulated expression of Bcl-2 m RNA, h TERT m RNA and up-regulated expression of caspase-3 m RNA.
