ObjectiveBy observing the expression of parafibromin in head and neck squamous cell carcinoma(HNSCC),we try to explore the relationship between which clinicopathological features and prognosis.Methods1.The wax tissues which confirmed with normal oral mucosa, oral mucosa dysplasia and HNSCC were selected accurately.We collected the Pathological information of all these patients and followed up with them.2.To observe the expression of parafibromin in tissue samples through tissue microarray technique and immunohistochemical method.3.Data were performed with SPSS21.0 Software,and statistically significant level was considered as P<0.05.Spearman rank correlation coefficient test was used to analyze the relevant data.Establishing the survival curve with Kaplan-Meier to describe the relationship between parafibromin-expressing and survival rate of HNSCC patients. Using Cox proportional hazards regression to establish the model of the data.ResultsThe expression of parafibromin detected in the nucleus of normal oral squamous epithelial cells and dysplastic cells was significantly higher than those in both primary and metastatic HNSCC cells.Parafibromin expression was negatively correlated with lymph node metastasis and TNM staging, but positively with HPV positivity and no no significant correlated with other data.The survival curve analysis showed that there was no significant difference about survival rate between the positive and negative patients in parafibrominexpressing(P=0.319).But the recurrence free survival rate of the positive patients of parafibromin- expressing is higher than other groups(P=0.043).Multi-factor analysis with Cox proportional hazards model shows that the distant metastasis of the tumor was an independent prognostic factor for overall survival( P=0.015),while the expression of parafibromin was an independent prognostic factor for relapse free survival(P=0.025).ConclusionDown-regulated expression of parafibromin may participate in the process of HNSCC initiation as an advanced marker.The expression of parafibromin may paly an important role in the molecular basis of HNSCC progression and metastasis.We may consider parafibromin expression was a molecular marker for tumorigenesis、metastasis and progression of HNSCC.
