Objective:Through observing the protective effect of total flavonoids from Taraxacum mongolicum on leptomeningeal microcirculation of mice, small, cerebral ischemia reperfusion rats model.Through the change of total flavonoids of Herba Taraxaci on blood flow to the brain in mice, the content of LDH cerebral ischemia reperfusion model of the small rat brain tissues (LDH), ATP enzyme, nitric oxide synthase (NOS), superoxide dismutase (SOD) activity, lactic acid (LD), nitric oxide (NO), malondialdehyde (MDA), interleukin-1β(IL-1β), Cell adhesion factor 1 (ICAM-1), vascular cell adhesion factor IVCAM-1, tumor necrosis factor-a(TNF-a) and the content of neuron specific enolase(NSE),the protein of S-100β(S-100β) MPO level in serum and pathological changes of cortex and hippocampus areas,so as to investigate the protective effect of cerebral ischemic reperfusion injury changes, total flavonoids from Taraxacum mongolicum on mice cerebral blood flow of small rats.Methods:Establish the meningeal microcirculation model in mice by ligating bilateral common carotid artery. Before operation, the corresponding group of mice were given nimodipine, Naoluotong, high, middle, low dose of dandelion flavonoids suspension gavage, the blank and model group were fed with the same amount of sodium carboxymethyl cellulose (0.5%CMC),1 times a day,7 days in a row. start test after the last administration of lhour (fasted 12h). Cut tie volume perfusion before 1 minutes 50 seconds to --2 minutes average as a section bar before the average perfusion, take blood perfusion of 3 minutes and 50 seconds to --4 minutes average as a sectional bar after the average perfusion, After ligation of the blood flow perfusion with average minus the amount of blood flow perfusion before ligation average divided by the average blood flow perfusion before ligation, and thus calculate the cerebral blood flow in mice modeling decline rate.Firstly bilateral common carotid artery ligation, ischemia lOmin, reperfusion 10min,then ischemia10min complex,the mice model of cerebral ischemia-reperfusion were found.Before the operation, the corresponding group of mice were given nimodipine, Naoluotong, high, middle, low dose of dandelion flavonoids suspension gavage, the blank and model group were fed with the same amount of sodium carboxymethyl cellulose (0.5%CMC),1 times a day,7 days in a row. In the last time after feeding 1h,24h after operating, half an hour later abstracting the eye blood, setting aside for half an hour,after centrifugation, the supernatant was measured the content of NSE, S-100β, the Brain tissue homogenatecontent of mice was measured SOD, LDH, ATP enzyme activity and MDA, LD and protein, observed morphological changes of mouse brain tissue.To establish the focal cerebral ischemia reperfusion rats model by blocking the left middle cerebral artery. Before operation, the corresponding group of mice were given nimodipine, Naoluotong, high, middle, low dose of dandelion flavonoids suspension gavage, the blank and model group were fed with the same amount of sodium carboxymethyl cellulose (0.5%CMC),1 times a day,7 days in a row. In the last time after feeding 1h,22h after operating, half an hour later abstracting the eye blood, evaluated neural function deficiency mark,measure the activity of ATP enzyme and the content of NO, NOS, IL-1β,ICAM-1, VCAM-1,TNF-α,the protein in brain tissues and the content of NSE and MPO in serum and observed morphological changes of mouse brain tissue.Results:The model of the brain microcirculation of mice,Cerebral Ischemia-reperfusion in mice repeatedly and the partial cerebral ischemia-reperfusion model in rats were duplicated successfully. Total flavonoids from Taraxacum mongolicum in each dose group promoted the brain microcirculation of mice significantly(P<0.01), through improving mean blood perfusion and cerebral blood flow after coronary artery ligation;reduced mortality significantly and neural function score of mice; Large doses of dandelion flavonoids group can make NSE、S-100β and MPO level in mice and rat serum,were lessened significantly (P<0.01), middle doses of dandelion flavonoids group can make NSE、S-100β in rat serum decrease obviously (P<0.05), low doses group of dandelion flavonoids group can reduce the trend of NSE、S-100βin serum (P) 0.05), can make MPO level in rats serum decrease obviously (P<0.05), small doses of dandelion flavonoids group can make the NSE level in serum was significantly decreased (P<0.01); Big, in, small doses of dandelion flavonoids group can make MDA, IL-1β, ICAM-1, TNF-a, NOS, LD and LDH activity decrease in brain significantly (P<0.01); high, medium doses of dandelion flavonoids group can make SOD activity in brain tissue of mice and ATP enzyme in mice and rats increased significantly (P<0.01); small doses of dandelion flavonoids group can make Mg++-ATP enzyme, Ca++-ATP enzyme activity in the brain tissue of mice was significantly increased (P<0.01), Na+K+-ATP enzyme activity was obviously increased (P<0.05), Na+K+-ATP enzyme, Mg++-ATP enzyme activity was raised significantly (P<0.01), Ca++-ATP enzyme activity in rats was obviously increased (P<0.05); Large doses of dandelion flavonoids group can make the NO, VCAM-1 level in rats brain tissue was significantly decreased (P<0.01), middle doses of dandelion flavonoids group can make NO in brain tissue of rats was decreased significantly (P<0.01), VCAM-1 were decreased obviously (P<0.05), small dose of dandelion flavonoids group can make the NO and VCAM-1 levels in brain tissue of rats decrease obviously (P<0.05); improve the injuries of hippocampus,cortex of the brain tissues(P<0.01).Conclusion:The total flavonoids of dandelion can protect the tissue injury of brain,through improving the mice collateral circulation and local blood circulation after the model, raising blood flow, lessening the blood flow rate of decline, thereby improving the meningeal microcirculation in mice; activating LDH, SOD and ATP enzyme of the brain; improving brain energy supply disturbance; reducing brain injury; reducing the content of LD, NOS activity and NO level, IL-1β, ICAM-1, VCAM-1, TNF-α and the content of MDA in brain tissue; reducing the content of NSE, MPO, S-100βin serum; lessening mortality of mice; improving the nerve function defect score; improving the pathological changes of hippocampus and cerebral cortex,thereby reducing brain ischemia reperfusion injury.
