Purpose: Through the observation of spontaneous hypertensive rat (SHR) expressionand the pathological changes and vascular extracellular matrix organization structure, aims toclarify the vascular remodeling and vascular function and correlation of the micro expressionof inflammatory factors, and to study the Xifeng Jiedu Tongluo Decoction intervention effecton high blood pressure of vascular remodeling, and to explore its mechanism of action fromthe cell the molecular level. In order to better TCM treatment of hypertension disease play arole, to improve the curative effect and provide a theoretical basis.Method: The morphology, immunohistochemistry, radioimmunoassay, molecularbiology methods, starting from the animal experiments, WKY rats were used as control, thedetection of spontaneously hypertensive rats (SHR) by different doses of Xifeng JieduTongluo Decoction (high, low dose group) and Western medicine group with benazepril aftertreatment, impact index matrix and protein expression of extracellular: observe the aortictissue of pathophysiology; Study on Xifeng detoxification Huoluo Decoction on vascularremodeling of hypertension intervention effect of Xifeng Jiedu Tongluo Decoction;spontaneously hypertensive rats (SHR). The mechanism of ECM synthesis and degradationmechanism and vascular remodeling.Result: In the drug intervention after eight weeks, the western medicine group andXifeng Jiedu Tongluo Decoction intima-media thickness (M) of the high dose group weresignificantly lower than that of model group, and the difference was statistically significant(P<0.05); in addition, the western medicine group and Xifeng Jiedu Tongluo Decoction highdose group, middle thickness/lumen diameter (M/L) was significantly lower than that ofmodel group, and the difference was statistically significant (P<0.05). High dose of XifengJiedu Tongluo Decoction intervention also make SHR rat carotid arterial pressure decreasedsignificantly than that in model group (P<0.05). Compared with the model group, XifengJiedu Tongluo Decoction high dose group ET-1, type I, III collagen, the expression level ofMMP-9decreased significantly (P<0.05); while the NO metabolite, TIMP-1expression wassignificantly increased (P<0.05). Conclusion:(1) Xifeng Jiedu Tongluo Decoction can significantly decrease the serumET-1content, and increased the activity of NOS in aorta tissue, suggesting that Xifeng JieduTongluo Decoction can improve vascular endothelial dysfunction (2), recovery of NO/Ang â…¡balance, improve endothelial function, so as to maintain the stability of the structure andfunction of blood vessels from the body. In addition, the study also found that in the modelgroup MMP-9, TIMP-1, I, collagen type III was significantly higher than that in normal group;compared with the model group, Xifeng Jiedu Tongluo Decoction group and medicine groupdecreased; correlation analysis showed highly positive correlated with MMP-9, TIMP-1, I,collagen and other indicators, said the close relationship between the Ming ECM synthesisand the degradation and vascular remodeling in spontaneously hypertensive rats, Xifeng JieduTongluo decoction may be one of the mechanisms of vascular remodeling in spontaneouslyhypertensive rats.
