Objective The aim of this study was to explore the effect of crude realgar、purifiedrealgar and the supernatant on the methylation status of socs-1gene in multiple myelomacell lines KM3.Methods The cell viability was assayed by MTT method.Morphological assessmentof apoptosis was performed with Wright-Gimsa staining,The cell apoptosis was analyzedby flow cytornetry. The methylation status of socs-1gene was detected by MSP.Results The results indicated that hyper methylation of CpG island of socs-1genewas observed without expression of socs-1in myloma cell lines KM3. The degrees ofmethylation was Observed significantly reduced or disappeared after exposure to cruderealgar for48hours as compared with purified realgar. The MTT assay showed that cellproliferation was inhibited in a concentration-dependent manner in crude realgar-treatedKM3cells. The apoptosis rates for20、30、40mg/l crude realgar after48h in KM3cellswere (43.90±2.62)%(52.80±1.87)%(54.00±3.28)%respectively in KM3cells. With thecontrol group(4.14±0.11)%,there were significant differences(P<0.05).The MTT assayshowed that cell proliferation was inhibited in a concentration-dependent manner inpurified realgar-treated KM3cells. The apoptosis rates for30、40、50mg/l purified realgarafter48h in KM3cells were (54.14±2.09)%(55.47±1.81)%(75.38±2.06)%respectively in KM3cells. With the control group(4.14±0.11)%,there were significantdifferences(P<0.05).Conclusion It is concluded that crude realgar may induce socs-1demathylation andup-regulate the expression of the gene.This study provides a new thought and direction forexploring possible mechanism of cell apoptosis by crude realgar and multiple myelomaby crude realgar.
