Objective: To observe the growth status of nano realgar, on human breast cancer cellline MCF-7cells of epithelial mesenchymal transition (EMT) affect regulation factorexpression and cell invasion, mechanism of nano realgar inhibited EMT in anti metastasis.Methods: Breast cancer MCF-7cells were cultured in vitro, dosing according toexperimental group, to observe the changes in cell morphology and growth condition,scratch test, Transwell invasive assay cell migration and invasion in change detection,immunohistochemical method to test the expression of E-cadherin protein(E-cad) andHypoxia inducible factor-1alpha(HIF-1α),Snail expression was detected by mRNART-PCR method.Results: Realgar nanoparticles can inhibit the growth of human breast cancer MCF-7cells, upregulation of E-cad, down regulated the expression of HIF-1α, SnailmRNA inMCF-7cells, can reduce cell invasion, migration, which were positively associated withthe drug concentration, and synergistic effect of combined with doxorubicin.Realgar nanoparticles can inhibit the growth of human breast cancer MCF-7cells, anddecreased cell migration, invasion, could up regulate the expression of E-cad, downregulation of HIF-1α, SnailmRNA expression in MCF-7cells, and its role in the drugconcentration was linear, and synergistic effect of combined with doxorubicin.Conclusion: Realgar nanoparticles can play the anti tumor invasion and metastasis byinhibition of EMT tumor cells.
