Objection: According to observe and study the expression of the Matrixmetalloproteinase-9(MMP-9) and the Tissue inhibitor of metalloproteinase1(TIMP-1) in lung tissues of pulmonary fibrosis rat models by giving ChaiHudrain-dampness decoction, to explore its mechanism in the treatmentof pulmonary fibrosis, and to provide experimental basis on the clinicalapplication for traditional Chinese medicine in the treatment of pulmonaryfibrosis.Methods:90healthy male Wistar rats, weight200g±20g, were divided randominto5groups: blank control group,model group,prednisone group,ChaiHudrain-dampness decoction group, ChaiHu drain-dampness decoction andprednisone group,18rats in each group. All of groups were injected thebleomycin by trachea to set up the model of pulmonary fibrosis, except blankcontrol. When the14th and28th day, Dynamically contrast the path-morphologychange and the protein expression of the MMP-9、TIMP-1of rats lung tissuein every groups. By applying SPSS19.0to analyze the experimental data andstatistical processing.Results:(1) At the14th day, the model control group rats had alveolitis inlung tissues and at the28th day, degree of pulmonary fibrosis is obvious, meanwhile, the alveolitis and pulmonary fibrosis degree in all the treatmentgroup rats on each time point was lighter compared with model control group.(2) At the14th day, the expression of MMP-9in rats’ lung tissue had a smallamount in model control group, at the28th day, the expression of MMP-9inrats’ lung tissue has decreased, but both were significantly higher than thatof normal control group. At the14th and the28th day, in ChaiHu drain-dampnessgroup、prednisone group and ChaiHu drain-dampness combine with the prednisonegroup, the lung tissue expression of MMP-9is less than the model controlgroup, compared with model group, the difference was statisticallysignificant (p <0.05).(3) At the14th day, the expression of TIMP-1in rats’lung tissue had a small amount in model control group; at the28th day, theexpression of TIMP-1in rats’ lung tissue has increased. Compared with normalcontrol group, difference was statistically significant (P <0.05). At the14th day,in ChaiHu drain-dampness group and prednisone group as well as ChaiHudrain-dampness combine with the prednisone group, the expression of TIMP-1in rats lung tissues is less than the model control group, comparedrespectively with model group, the difference was statistically significant(p<0.05); At the28th day, each treatment group the expression of TIMP-1inrats’ lung tissue were increased, compared respectively with model group,the difference was statistically significant (P<0.05).(4)At the14th andthe28th day, the expression of MMP-9and TIMP-1in rats’ lung tissue inthe prednisone group and ChaiHu drain-dampness decoction group had nodifference in statistical significance (P>0.05); While compared with theChaiHu drain-dampness combine with the prednisone group, the two kinds of testindex difference was statistically significant (P <0.05).Conclusion:(1) In the early pulmonary fibrosis, the lung tissue expressionof MMP-9and TIMP-1were increased, but with MMP-9increased obviously; Andin the late period, pulmonary fibrosis MMP-9protein expressions was decreased,While the expression of TIMP-1was continuous high. It suggested that, during early pulmonary fibrosis, MMP-9had a high expression, and in the late periodwas reduced, leading the ECM increased maybe one of the reasons about theformation of pulmonary fibrosis.(2) ChaiHu drain-dampness decoction cansignificantly reduce and the general performance of pulmonary fibrosis modelrats lung tissue pathology change, indicating it have anti-fibrosiseffect.Speculated that the ChaiHu drain-dampness decoction can treatpulmonary fibrosis by decreasing the expression of MMP-9in early period andrestrain the expression of TIMP-1in late period to adjust the metabolism ofECM. And the treatment effect on ChaiHu drain-dampness decoction combined withthe prednisone is superior to the single drug use.
