| bjective:To investgate the effects of oral administere-d icariin and breviscapine on intracavenosal pressure(ICP)/mean ar-terial pressure(MAP) and on expression of the endothelial nitric oxi-de synthase （eNOS）,neuronal nitric oxide synthase（nNOS）,Pho-sphodiesterase type5(PDE5), Rho kinsase（Rock1、Rock2）in corp-us cavernosum of spontaneous hypertensive rat (SHR). Methods:5WKYs and20SHRs（12weeks old） were randomly divided into5groups of5SHRs or WKYs:WKY control group（A）,SHR cont-rol group（B）,icariin–treated group （C）,breviscapine treated gro-up （D）,drug combination–treated group （E）. The groupA and Bwere treated with double distilled water;and the groupC were tre-ated with icariin（10mg/kg）;groupC were treated with breviscapine-(80mg/kg),groupC were treated with icariin（10mg/kg） and brevisc-apine(80mg/kg) orally for4weeks.The changes of ICP/MAP obser-ved continuously,then the serum testosterone were detected. The ex-pressions of Enos,nNOS，PDE5，Rock1，Rock2were analyzed byWestern blot and immunohistochemistry. Results:the body weight（g-roupA:278.8±8.5g,groupB:275.3±4.5g.groupC:283.2±2.1g.groupD:274.0±2.8g.groupE:277.4±3.5g）and serum testosterone（groupA:400.7±18.20ng/dl.groupB:475.11±34.33ng/dl.groupC:489.66±60.95ng/dl.g- roupD:428.42±46.60ng/dl.groupE:483.11±42.33ng/dl） had not si-gnificantly difference in each group，the MAP in group A（90.75±6.54mmHg） was significantly lower than the other groups (groupB:173.72±6.23mmHg.groupC:180.45±5.62mmHg.groupD:181.52±6.73mmHg.groupE:179.43±6.25mmHg)(P <0.05);By measuring eac-h group ICP/MAP at3Vor5V voltage stimulation,group A (0.75±0.056,0.81±0.083)was significantly higher compared with other gro-ups (gro-upB:0.21±0.021,0.026±0.026;groupC:0.30±0.024;0.33±0.021;groupD:0.38±0.088,0.46±0.038;groupE:0.43±0.132,0.59±0.074)(P <0.05);group C(0.30±0.024；0.33±0.021) and group D(0.38±0.088,0.46±0.038) was significantly higher than group B (0.21±0.021,0.026±0.026)(P <0.05), group E (0.43±0.132,0.59±0.074)was sig-nificantly higher than groupB(0.21±0.021,0.026±0.026),C(0.30±0.024;0.33±0.021）,D（0.38±0.088,0.46±0.038）, group D（0.38±0.088,0.46±0.038） was significantly higher than group B (0.21±0.021,0.026±0.026)(P <0.05); Immunohistochemistry showed eNOS ismainly expressed in the vascular endothelial cell membrane，nNOSis mainly expressed in neurons， PDE5, Rock1andRock2were ma-inly expressed in the cytoplasm of smooth muscle cells；Western b-lot showed that eNOS （0.129±0.011）and nNOS （0.101±0.014）of Group A was significantly increased compared with the other gr-oups（Enos:groupB:0.032±0.0019,groupC:0.054±0.0031,groupD:0.040 ±0.0024,groupE:0.104±0.0062;nNOS:group B:0.054±0.0046,groupC:0.070±0.0060,groupD:0.057±0.0048,groupE:0.088±0.0074）(P <0.05),group-C （0.070±0.0060）compared with group B（0.054±0.0046） was significantly increased (P <0.05),group E（0.104±0.0062）was significantly higher than the group B（0.054±0.0046）,C（0.070±0.0060）,D（0.057±0.0048）(P <0.05);the PDE5of group A(0.087±0.0034) and E(0.094±0.0066) were significantly decreased co-mpared wit-h group B(0.168±0.012) and D(0.141±0.0099)(P <0.05),group C(0.1±0.0071) compared with group B(0.168±0.012) wassignificantly lower (P <0.05),the Rock1of group A(0.025±0.0015)were signif-icantly decreased compared (P <0.05)with groupB(0.075±0.0032),C(0.074±0.0025) and D(0.052±0.0018),compared with gr-oup B (0.052±0.0018)and C(0.074±0.0025),group D(0.052±0.0018)was signify-cantly decreased (P <0.05);the Rock2of group D（1.024±0.027）a-nd E（0.888±0.023）were significantly lower than gro-upB（3.142±0.414）and C（1.516±0.027）,respectivetly(P <0.05),g-roup C(0.074±0.0025) compared with group B(0.075±0.0032) wassignificantly lower (P <0.05),group A（0.622±0.026) was significan-tly lower th-n the other groups (P <0.05). conclution:The combin-ation treatment with breviscapine and icariin has been significantly i-mproved erectile function in SHR,showed that combination therapy-can be used as a new method for the treatment of hypertension c- aused erectile dysfunction. |
