Objective: To detect Kazakh esophageal squamous cell carcinoma (ESCC) and adjacent normal tissues(NETs) PLCE1, PKCα proteins and NF-κB related proteins expression, analyze the relationship betweenthe clinical and pathological features, and the correlation between them.Methods: A case-control study, select90cases with Kazakh ESCC (case group) and48cases for normal(control group), detect ESCC tissues with PLCE1, PKCα proteins and NF-κB relate proteins, withimmunohistochemical (IHC) of Envision or SP methods. Using χ2test to analyze whether there aredifferences between the ESCC and NETs, and relationships between expression and clinicopathologicalfeatures. Among the various proteins, using Spearman’s rank correlation further analysis of the correlationsbetween them.Processing applications with SPSS13.0software.Results: The high expression rates of PLCE1,PKCα, IKKβ, IKBα, p50and p65proteins in ESCC78.96%(71/90),45.6%(41/90),53.3%(48/90),40%(36/90),81.1%(73/90)and56.7%(51/90)respectively,were higher than those expression in NETs35.4%(17/48),8.3%(4/48),22.9%(11/48),16.7%(8/48),47.9%(23/48) and37.5%(18/48) respectively, all P<0.05. The expression of PLCE1protein wasoverexpression in ESCC with lymph node metastasis90.2%(46/51) and TNM early stage93.3%(28/30),PKCα protein with poor differentiation54.7%(35/64), IKKβ and IKBα proteins with TNM stage66.7%(40/60)、56.7%(17/30), p50with lymph node metastasis88.2%(45/51)and p65with depth of tumorinfiltration70.5%(31/44). PKCα, IKKβ and p50proteins expression positive correlation with PLCE1(PKCα:r=0.259,P=0.017;IKKβ: r=0.246, P=0.017; p50: r=0.244, P=0.024); the mean histoscore ofIKKβ, PKCα and p50expression were higher in ESCC tissue with high levels of PLCE1expressionrespectively,which IS (Immunohistochemical score, IS) were (8.258±3.066,7.250±3.396and8.667±3.038respectively) than in those with low levels of PLCE1expression which IS (6.613±3.584,5.750±3.317and6.407±3.479respectively (all P <0.05).Conclusions:(1) PKCα, PLCE1proteins and NF-κB correlated proteins IKKβ, IKBα, p50and p65wereoverexpression in Kazakh’s ESCC and correlated with poor prognosis.(2) PKCα, PLCE1proteins andNF-κB signaling pathway synergy promote Kazakh’s ESCC.(3) PLCE1—PKCα—NF-κB signalingpathway in xinjiang kazak’s ESCC may be activated, which provide molecular targets for treatment andprevention of Xinjiang kazak’s ESCC.
