Objective:Isolated of the acetone extract of Turkish gallsget different preparativefraction by Polymer materials MCI and ODS. Discussion and Studyof Turkish gall extract part of anti-radiation by Modern biology technology, Identifyanti-radiation mechanism. Methods:1、Isolated of the acetone extract of Turkish gall ofMCI methanol water system by column chromatography, Isolated of theMCI extract ofTurkish gall with ODS methanol water system by column chromatography. With the gallicacid as reference, total tannins and gallic acid of Turkish galls and its different preparativefraction were determined by high performance liquid chromatography and ultravioletspectrophotometer, respectively.2、 Because lymphocytes are extremely sensitive toradiation, Establish a one-time radiation damage model with8Gy X ray irradiated byhuman lymphoblastoid cells (AHH-1), the experimental group divide into non-controlgroup, model group,5μ g/ml,25μ g/ml,50μ g/ml,100μ g/ml,200μ g/ml MCI extractgroup, selection of the best group in anti-radiation by MTT. The group detect cell cyclesby PI staining, Annexin V/PI stainingis used to detect apoptosis rate, the level of ROSfluorescence probe DCFH-DA detection in cells, Western blot were mediated by themitochondrial pathway of apoptosis and caspase-3family signaling pathway change,affect the expression of Bax, Bcl-2, Caspase-3, JNK, ERK, p-ERK, p-JNK protein.Results:1The difference between extracts of gallic acid content significantly, especiallyin the ODS water extract content reached54.6%, Turkish galls content5.76%, ODS waterextract is9.48times Turkish galls of gallic acid. The total tannin content of differentextract parts (except ODS water extract) than the Turkish galls content increasedsignificantly, the highest to40%MCI of methanol is63.53%, Turkish galls is42.85%. Thedetermination results of gallic acid and total tannin content, combined with dry powder ofdifferent extracts yield condition, study on anti radiation activity cytology choice40% methanol MCI extracts.2Radiation can induce AHH-1apoptosis.5μg/ml and25μ g/mlgallic MCI extract group can improve the survival rate of X ray radiation of AHH-1, withthe most obvious effect of5μg/ml group (P<0.01).5μg/ml gallic MCI extract cansignificantly reduce the radiation effects of G1cell cycle arrest, on radiation induced cellsin S phase arrest in G2phase delay, also have certain effect. Cells of early, late apoptosis5μ g/ml gallnut extract can inhibit the MCI radiation induced (P<0.01).5μg/ml showedsignificantly reduced radiation effect on cellular ROS levels (P<0.01), gallic MCI extractcell ROS level was model group11.43%(P<0.01). Conclusion: The effective componentcontent determination analysis showed that MCI or ODS column chromatography ofacetone extracts of Galla, it provides a new idea for gall medicine in the full use of,improve efficacy, monomers etc.Low dose (5μg/ml) gallic MCI extract has obviousimprove the survival rate of cells of X ray irradiated AHH-1, alleviate the cells G1phasearrest, decreased cell apoptosis, reduce reactive oxygen species generated by radiationeffect. Anti radiation effect and possible mechanism of gallic MCI extract antioxidanteffects, can maintain the balance and stability of the original passage cells, mitochondriainduced by radiation, the release of cytochrome C and the pro apoptotic Bax protein,inhibit the apoptosis induced by MAPK signal pathway. Studies show that gallnut extractpart has good radiation protection effects, and lay the foundation for the development ofnew natural radiation protection medicine.
