Objectives:Moxibustion therapy on experimental RA model to observe the anti-inflammatory and detumescence and explore moxibustion on RNA interference rheumatoid arthritis rabbits’s serum cytokines IL-17, MMPs and its anti-inflammatory effect mechanism.Methods:Japanese white rabbits were randomly divided into control group, model group, moxibustion group, moxibustion+SOCS gene interference group (referred to SOCS interference group), moxibustion+PTPN gene interference group (referred to PTPN interference group), moxibustion+PIAS gene interference group (referred to PIAS interference group), n=6. Injected freund’s complete adjuvant by0.5ml/Kg average into the rabbit’s double rear knee joint cavity to shape RA model. Used microinjector to inject lentiviral vector mediated siRNA40μg/each one average into the gene interference rabbit’s double rear knee joint cavity respectively on the next day and moxibustion treatment tenth days.Moxibustion group and moxibustion+gene interference group were treated by moxibustion moxa sides "shen" points each five Zhuang,1day,6d for a course of treatment,three courses in all. Immune ELISA (Elisa method) were used in strict accordance with operating procedures to determine in each experimental group rabbit serum IL-17,MMPs levels after the treatment.Results:1.Compared with the blank group,the perimeters of bilateral knee joints were significantly increased in the model group (P<0.01); compared with the model group, the perimeters of bilateral knee joints were significantly decreased in the moxibustion group (P<0.05); The perimeters of bilateral knee joints in SOCS interference group, PTPN interference group and PIAS interference group decreased less than moxibustion group (P<0.05).2.Compared with the control group, model group rabbit serum IL-17levels were significantly higher (P<0.01); Compared with model group, moxibustion group rabbit serum IL-17levels have decreased significantly (P<0.01); PIAS interference group and SOCS interference group serum IL-17levels were significantly lower (P<0.05), PTPN interference group tended to decrease; PTPN interference group, PIAS interference group and SOCS interference group reduced IL-17levels less effective than moxibustion group。3.Compared with the control group, model group rabbit serum MMPs levels were significantly higher (P<0.01);Compared with model group, moxibustion group, PTPN interference group, PIAS interference group and SOCS interference group rabbit serum MMPs levels have decreased significantly (P<0.01); PTPN interference group, PIAS interference group and SOCS interference group reduced IL-17levels less effective than moxibustion group.Conclusions:1.Moxibustion with anti-inflammatory, detumescence effect on experimental RA.2.Moxibustion’s anti-inflammatory effects on experimental rabbits may because of reducing cytokine IL-17and MMPs levels.3.One of the important mechanism of Moxibustion in treating RA may be related to effects of negative feedback factor SOCS1, PTPN22, PIAS1to produce positive role in the regulation of the JAK-STAT pathway, and down-regulation of IL-17, MMPs.
