| Bletillae Rhizoma (Bai-Ji in Chinese), the rhizoma of Bletilla striata (Thunb.)Reichb.f., is distrbuted widely in South and Southwest China. Bletillae Rhizoma is wellknown as one of key herbs for stoping bleeding and promoting tissue regeneration, andfrequently applied for curing gastrointestinal ulcers and bleeding in Chinese TraditionalMedicine. Recent investigations suggested that polysaccharide of Bletilla striata (bletillan)had contributed to pharmacological effects of the herb. Quality standard of BletillaeRhizoma has been recommended in Chinese Pharmacopeia (2010edition), butquantification of bletillan remains not established yet. In this thesis, a method wasdeveloped for determination of bletillan, and quality standard of Bletillae Rhizoma wasthen developed and recommended as an amending draft for the coming ChinesePharmacopeia (2015edition).A colorimetry method was established and verified for determination of bletillan atfirst, using anthrone-sulfuric acid reagent. Sample procedure was explored in detail anddeveloped as follows: herbal powders (0.5g) was weighed accurately, extracted with water(150ml) under reflux for1h, filtrated, and transferred to an volumetric flask (250ml);after filtration,0.5ml of which was accurately transferred to a tube, then added withacetone (4ml), centrifuged, the precipitate was solved with water and then transferred toanother volumetric flask (10ml) to give final sample solution. Using this methoddeveloped,12representative herbal samples were then analyzed. As a result, bletillancontents of samples of crude medicines and decoction pieces (6batches in each group)were24.12-35.78%(28.6%in average),20.52-28.38%(25.3%in average), respectively.Since the herbal medicine resources are limited, the limit of bletillan content of crudemedicine and decoction piece was recommended as17%and14%, respectively.Quality analysis of these12batch samples were then conducted according to standardof Chinese Pharmacopeia (2010edition), including characteristic identification,microscopic identification and thin layer chromatogram identification, water content andtotal ash content. As for thin layer chromatogram identification, methylene dichloridecould be applied as a substitute extraction solvent instead of diethyl ether during sample process. With quantification method and suggested content limit of bletillan and adjustedsample process method for thin layer chromatogram identification as mentioned before, anamending draft was finally recommended as quality standard of Bletillae Rhizoma infuture Chinese Pharmacopeia (2015edition). |
