Objective:To research whether styrax belongs to pregnancy contraindication, or it added to compound can treat dysmenorrhoea related to styrax, and to provide pharmacological evidence for rational drug use of clinical gynecologic.Methods:①To observe the effect of styrax on non-pregnant, early-pregnancy uterus in vivo and in vitro, to observe the interventional mechanism of styrax to resist erethism caused by oxytocin(OT) and acetylcholine(Ach)②To observe the effect of styrax on estrous cycle, level of E2and P, wet weight of reproductive organ in vivo.Result:(1) Effection and mechanism on uterine smooth muscle of nonpregnant rats and early pregnant rats in vivo:①The doses of Styrax0.67,1.34g/kg were chronically administered to rats for10days could remarkably increase the tension, peak area, frequency of non-pregnant rats uterus(P<0.05, P<0.01)②Given Styrax to the early-pregnant rats by gavage for3days showed no significant difference; The findings of duodenal administration shows:Styrax0.67,1.34g/kg were given by duodenal administration, could remarkably inhibit the tension and peak area, the frequency of the uterus(P<0.05, P<0.01).③Styrax1.34g/kg could obviously inhibit the exciting of the nonpregnant rats and early pregnant uterus caused by OT(P<0.01), it showed no significant difference by given OT again.(2) Effect on the estrous cycle and serum sex hormone:Styrax0.67,1.34g/kg were chronically administered to rats for10days, it could not only obviously prolong estrous cycle and lower the level of E2and P but also decrease the index of uterus and ovary (P<0.05,P<0.01).(3) The influence and the mechanism of Styrax on uterus of non-pregnant and early pregnant in vitro:①Styrax1.13mg/mL can obviously against the contraction of non-pregnant uterus induced by OT (P<0.01), given OT again, it obviously reinforce the frequency (P<0.05), but it made no difference to uterus after given Ach again; Styrax1.13mg/mL and atropine had synergistic inhibitory effects on uterus(P<0.05), there’s no difference on uterus by given Ach again after Styrax.②Styrax1.13mg/mL could obviously resist the excitatory contraction of early pregnant uterus caused by OT and Ach in vitro(P<0.01), added OT and Ach again made no remarkable difference; atropine didn’t showed significant inhibitory effection to uterus of early pregnant rats in vitro, Ach could exciting the tension,peak area, frequency that treated by Styrax P<0.05, P<0.01).Conclusion:①Continuous use high dosage of Styrax could significantly lower the level of E2and P in serum, intervene the estrous cycle and affect the development of reproductive organs, it’s warning high dosage and continuous long time use of Styrax or it’s preparation is unfavorable for female patients②Styrax show significantly inhibitory effect on uterus of nonpregnant and early pregnant rats, the mechanism may be related to blocking oxytocin receptor and M acetylcholine receptor partly. we can infer that Styrax’s complex prescription have the effection of treating dysmenorrhoea, it may be related to inhibit the anomaly contraction of uterus smooth muscle.
