Objective Observation with liver qi effect of flavored si-ni-san on nonalcoholic fatty liver rat liver inflammatory reaction related to endoplasmic reticulum stress, discuss the function mechanism of the prevention and treatment of NASH.Methods Using high fat diet for12Zhou build the NASH rats model, at the same time with6g/kg· d flavored si-ni-san intervention12weeks, oil red O staining using the detection of liver tissue fatty deposits;Conventional HE staining were liver tissue fatty change degree, the degree of inflammation;Biochemical method to detect liver tissue homogenate of TG,TC,ELIS A method to detect the serum TNF-a and IL-1β level, Western blot technique to detect liver tissue was detected in IRE la mediated inflammatory reaction of key molecular GRP78, XBP1signaling pathways, the Nf-kappa B, AP-1(C-Jun, C-Fos) protein expression.Results①Model group rats liver fat deposition, liver tissue fatty change degree, the degree of inflammation and NAFLD activity score (NAS) are significantly higher than normal group (P<0.01);The levels of serum ALT, AST, rats liver tissue homogenate of TG,TC content is significantly higher than normal group (P<0.05, P<0.01);Flavored si-ni-san party groups of rat liver tissue fat deposition, liver tissue fatty change degree, the degree of inflammation and NAS have dropped significantly in the model group (P<0.01);The levels of serum ALT, AST, rats liver tissue homogenate TG,TC content are different level lower in the model group (P0.05, P<0.01).②Model group rats liver tissue IREla mediated inflammatory reaction of key molecular GRP78,XBP1signaling pathways, the Nf-kappa B, AP-1(C-Jun,C-Fos) protein expression were significantly enhanced in the normal group (P<0.05,P<0.01), serum TNF-a and IL-1β level is significantly higher than normal group (P<0.05,P<0.01);Flavored si-ni-san party groups of rat liver tissue IREla mediated inflammatory reaction of key molecular GRP78,XBP1signaling pathways, the Nf-kappa B, AP-l(C-Jun,C-Fos)protein expression were decreased significantly in the model group (P<0.05, P<0.01), serum TNF-a and IL-1β level is significantly lower in the model group (P<0.05, P<0.01)Conclusion①ERS in NASH’s development induced by high-fat diet.②Flavored si-ni-san party group can inhibit the expression of ERS to regulate the liver lipid metabolic disorders, reduce the inflammation factor TNF-α,IL-1β, reduce liver inflammation, prevent the progress of NASH.
