Objective To explore the protective effects of dexmedetomidine on renalischemia-reperfusion-injury-induced kidney and liver injury in rats.Methods1.90male SD rats were randomly divided into3groups (n=30), pretreatedwith DEX group (DEX group), ischemia reperfusion group (IRI group) and controlgroup (Sham group). Rats were given DEX intraperitoneally100μg/kg in DEX group30min before surgey, the rest of the two groups were given the same amount of sterilesaline.2.A renal ischemia-reperfusion model was established, then the upper leftlobe of liver and the left kidney were harvested from rats in each group2h,8h, and24hafter reperfusion. Liver and kidney superoxide dismutase (SOD), nitric oxide (NO) andlactic acid (LD) and total antioxidant capacity (T-AOC) were detected2h,8h,24hafter reperfusion.3. Serum IL-17A and TNF-α changes of dexmedetomidine preconditioninggroup (DEX group), ischemia-reperfusion group (IRI group) and control group (Shamgroup) were measured by using a microplate reader2h,8h,24h after reperfusion.4. Serum creatinine (Cre), urea (Urea), alanine aminotransferase (ALT) ofthree groups of rats were observed by using a biochemical analyzer2h,8h,24h afterreperfusion.5. Changes in kidney and liver pathology of three groups of rats2h,8h,24hafter reperfusion were observed by HE staining.Results1. Compared with IRI group, kidney SOD and T-AOC increased in DEXgroup2h,8h,24h after reperfusion, and the total NO reduced (P <0.05), LD reducedsignificantly at8h after reperfusion (P<0.05).2. Compared with IRI group, liver SOD, total NO and T-AOC increased inDEX group2h,8h,24h after reperfusion (P<0.05), LD reduced significantly8h and24hafter reperfusion (P <0.05).3. Compared with IRI group, IL-17A levels reduced in DEX group2h,8h, 24h after reperfusion (P<0.05), and TNF-α levels were also lower than IRI group,while the value arrived the lowest point24h after reperfusion (P <0.05).4. Compared with IRI group, the concentration of ALT decreased2h,8h,24h after reperfusion (P<0.05) in DEX group, and the value of Cre reduced significantly8h and24h after reperfusion (P<0.05), and the concentration of Urea decreasedsignificantly8h after reperfusion(P <0.05).5. HE staining showed that, compared with the IRI group, edema of tubularepithelial cells was reduced in DEX group, consitent with the cytoprotection, DEXgroup had more normal histomorphology and less congestion of glomerular; liver cellsedema was relieved in DEX group, and liver cells arranged more orderly, and sinusoidaldilatation and congestion was eased, while inflammatory cell infiltration was alsoreduced.Conclusion Pretreatment with dexmedetomidine can enhance kidney and liverantioxidant capacity after renal ischemia reperfusion injury in rats, and reduce lacticacid accumulation.
