Objective:By research the effect of hand acupuncture and electroacupuncture on mechanical paw withdrawal threshold(MWT) and thermal withdrawal latency(TWL) in c-kit knock-out rats, to observe the relationship between mast cells and analgesic effect of acupuncture, to explore the mechanisms of the effect of pain threshold in mast cell deficient(c-kit knock-out) rats of acupuncture.Methods:C-kit knockout rats (WsWs-/-) and the wild contrast rats (WsWs+/+) of each30, two groups’control were tested in rats with MWT and TWL, then hand needle (MA),1mA electroacupuncture (1mA EA),3mA electroacupuncture (3mA EA) stimulated rat left Zusanli10min,10min later,test the pain threshold again.Results:1.Compared to c-kit knockout rats and wild contrast rats have increased in basic mechanical pain threshold.In the two groups,the MWT of right metapodium compared to the data was statistically significant (P<0.05),the left side have no statistical significance,but the MWT results of c-kit knockout rats compared with the control rats also tends to increase;There were no significant difference on TWL.2.The comparison of the MWT of time in WsWs+/+rats,Left of1mA、3mA EA and control compared with larger differences,3mA is better than1mA in it, P<0.001, P<0.01;Right of electroacupuncture and control compared with larger differences,There are differences between the hand acupuncture,EA is better than hand acupuncture,P<0.001,P<0.001,P<0.05.3.After three different stimulations, The MWT intensity of WsWs+/+rats are same with the MWT of time in WsWs+/+rats.4.The MWT intensity of WsW-/-rats compared with post-stimulate rats,There is no difference between the MWT of post-stimulation and control in left side;There are differences between the MWT of post-stimulation and control in right side,the effect of3mA is better than1mA,P<0.05,P<0.01.5.The MWT time of WsWs-/-rats compared with post-stimulate rats,The MWT time of WsWs-/-rats are same with the MWT of time in WsWs-/-rats.6.The TWL time of WsWs+/+rats compared with post-stimulate rats,There are big differences between the TWL of post-stimulation and control in left side,P<0.001;The same to the right metapodium,P<0.001, P<0.001, P<0.01.7.The TWL time of WsWs-/-rats compared with post-stimulate rats,There are big differences between the TWL of post-stimulation and control in left side,P<0.001;The same to the right metapodium,P<0.001.Conclusion:For the base pain threshold of WsWs+/+and WsWs-/-rats,WsWs-/-rats’ control is higher than WsWs+/+rats in WMT,but there is no difference in TWL between two groups.The effect of electroacupuncture on MWT is closely related with the integrity of the mast cells,the effect of hand acupuncture have little to do with the integrity of the mast cells.No matter what kind of stimulation caused by the change of TWL,there is no significant relationship between the TWL and the integrity of the mast cells.
