ObjectiveResearch on cancer stem cell surface markers CD44and proliferating cell nuclear antigen Ki-67expression and clinicopathological significance in laryngeal squamous cell carcinoma. and to observe and analyze the number of CD44+/Ki-67-cells in laryngeal squamous cell carcinoma, morphological characteristics and distribution relationships and clinicopathological parameters discussed CD44+/Ki-67-laryngeal squamous cell cancer stem cells as a marker of potential value.MethodsFrom General Hospital of Lanzhou Military Area Command Department of Pathology,20082012specimens of laryngeal surgery. screened69patients with squamous cell carcinoma. Another18cases of normal laryngeal mucosa tissues as controls. Using tissue microarray technology, immunohistochemistry staining and immunohistochemical double staining technique, positioning markers CD44+cells, Ki-67-cells and CD44-/Ki-67-cells, to observe and analyze their number in laryngeal squamous cell carcinoma, distribution methods, as well as the relationship between cell morphology and clinicopathological parameters. Using IBM SPSS20.0statistical package for statistical analysis, P<0.05difference was statistically significant.Results1. CD44+cells in the normal mucosa is almost non-existent, and in laryngeal squamous cell carcinoma is abundant quantity, the difference was statistically significant (P=0.000), similarly, Ki-67+cells in the normal mucosa only scattered basal cell layer, and in laryngeal squamous cell carcinoma abundant quantity, the difference was statistically significant (P<0.05).2. Patients with squamous cell carcinoma of the larynx, Ki67+did not differ by sex, age, clinical classification between CD44+cell count statistically significant difference (P>0.05), while the expression of the degree of differentiation between, T stage were significantly different (P <0.05).3. CD44+/Ki-67-cells in normal laryngeal mucosa almost no expression, only a small amount of CD44+/Ki-67-cells expression cancer tissue (1.23%and13.87%, average6.94%), Such cells have a more expression near in basement membrane in the vicinity, showing small focal or diffuse punctate distribution, mostly polygonal cells, projections and more, relatively little cytoplasm, nucleus round, deeply stained, are relatively immature cells differentiate.4. Indifference in gender, age, clinical type, CD44+/Ki-67-tumor cells express no statistically (P>0.05), while the expression of the degree of differentiation between, T stage and lymph node metastasis are statistically different.(P<0.05)Conclusions1. CD44-/K.i-67-cells in very small quantities in laryngeal squamous cell carcinoma differentiation relatively immature, which is closely related to the number and reflects the degree of malignancy and prognosis of the tumor clinicopathological parameters can be used as biological assessment of laryngeal squamous cell carcinoma behavior, determine the prognosis of one of the bases to provide reference for clinical treatment.2. In laryngeal squamous cell carcinoma, CD44T tumor cells in a small number of quiescent (GO phase), the majority of CD44-tumor cells in the proliferative phase (Gl, S, G2and M phases), laryngeal squamous cell carcinoma cancer stem cells is very GO phase may be present in CD44-tumor cells. CD44-/Ki-67-immune phenotype may be laryngeal squamous cell carcinoma cancer stem cell markers potentially valuable, is expected to become the next target for the treatment of laryngeal squamous cell cancer stem cell targeted therapy.
