Objective: to explore the expression and distributioncharacteristics of5-hydroxytryptamine receptor2B (5-HT2BR) in tumortissue, blood and gastric juice of patients with gastric cancer. Methods:1.80cases of paraffin specimens with gastric cancer tissue in our pathologydepartment were randomly sampled as the experimental group (n=30),according to the respective number from men and women during2006to2013.Half were male, the other half were women. Put paraffin specimensof gastric ulcer were removed surgically as control group during the sameperiod (n=10). There were no gastric cancer patients after chemotherapy,radiotherapy and immunotherapy before operation, detect the expressionof5-HT2BR by PV-9000immunohistochemical techniques, analyse therelationship among5-HT2BR and constituent ratio of patients, tumorstage, differentiated degree, histological type, infiltration depth andlymph node metastasis.2. Collect gastric juice and blood samples frompatients with gastric cancer those were determined by gastroscope andpathology detection(n=15)from October2013to January2014. Allpatients were admitted to surgical treatment. Establish the inclusioncriteria:(1)in preoperative diagnosis of gastric cancer by gastroscopy andpathological examination;(2)would do for gastric cancer radical surgery;(3)without gastric cancer that has spread to the liverand distant metastasis; (4)important viscera have good function, can tolerate surgery;(5)voluntary cooperate to clinical research. Exclusion criteria:(1)therecurrence of cancer, the repeated carcinoma, residual gastric cancer;(2)had a history of gastrointestinal surgery or gastrointestinal functionfailure;(3)immune function defects or combined with other diseases,such as, cholecystitis, appendicitis, systemic infection, etc;(4)function ofliver, kidney, heart and lung were not severely complete;(5)withhematopoietic system disease;(6)for severe malnutrition;(7)seriousdisorders about electrolyte and acid-base balance;(8)used ofglucocorticoid, Human blood albumin, immune preparation andtransfusion recently;(9)treated with preoperative chemotherapy, radiationtherapy, and other related. Collect gastric juice through the tube beforeand after the operation, extract of gastric juice in the operation by thestomach. Collected the peripheral blood (preoperative, intraoperative andpostoperative) and portal vein blood (intraoperative). Detect theexpression of5-HT2BR in the specimen after surgical by PV-9000.Collect gastric juice samples, blood samples (n=15) and gastric mucosalsamples (n=5) of patients with chronic gastritis, measure the expressionquantity of5-HT2BR using method of ELISA and PV-9000. Researchand analysis the expression situation of5-HT2BR in the patient’s blood,gastric juice and tumor tissue. Results:1. No positive expression of5-HT2BR in benign gastric tissues (n=15).2.Positive expression of5- HT2BR in gastric cancer tissue (n=42), positive rate is93.3%(42/45).Positive expressions are tan grains in gastric cancer cell cytoplasm.Staining in high differentiation of gastric cancer cell cytoplasm isrelatively shallow light, but colour is the deepest in poorly differentiatedcancer cell cytoplasm. The positive expression rate of5-HT2BR betweengastric cancer and gastric benign lesions has significant difference(P<0.01).3. Integral optical density value and positive area of5-HT2BRin low differentiation tissues of gastric cancer (G3) are higher than highand medium differentiation tissues of gastric cancer, the difference isstatistically significant (P<0.01), while the difference between highdifferentiation and middle differentiation tissues of gastric cancer is nostatistical significance.4. There is no statistically significant differenceamong5HT2BR and constituent ratio of patients, tumor stage,differentiated degree, histological type, infiltration depth and lymph nodemetastasis (P>0.05).5. There is no statistically significant differencebetween the blood and gastric juice of the patients with gastric cancer orpatients with chronic gastritis. But the differences about expression of5-HT2BR in the gastric juice and blood between gastric cancer patientsand chronic gastritis patients are statistically significant(P<0.01).Conclusion:1.5-HT2BR have high expression in blood, gastric juice andtumor tissues of patients with gastric cancer, not in gastric benign lesions.2. It is of great significance that detecting the expression of5-HT2BR in gastric juice and serum.3. The development of receptor blockers about5-HT2BR is expected to becomea new target.
