Objective: To observe the influence of high-fat diet on serum lipids,ALT,intestinalflora and inflammatory cytokines through feeding SD rats with high-fat diet.Meanwhile,toexplore the role of probiotics in regulating the intestinal flora and serum lipids throughobserving the impact of Saccharomyces boulardii on rats.Methods:40SD rats were randomly divided into four groups(10rats in each group).Group Awas the control group and was given normal diet. Group B was the high-fat dietgroup and was given high fat diet.Group C was the intervention group one and was givenSaccharomyces boulardii powder after10weeks besides high fat diet.Group D was theintervention group two and was given Saccharomyces boulardii powder from thebeginning besides high fat diet.To collect the feces and blood of the4groups’for testing atthe beginning of the experiment.Group C was given Saccharomyces boulardii by gavageuntil the end of the experiment after the feces and blood of the4groups’were collected fortesting at the tenth weekend.At the sixteenth weekend,to collect the feces and blood of the4groups’for testing again.Blood was used to testALT,TC,TG,LDL-C,HDL-C,LPS,TNF-α,IL-6.Feces was used to test the number oflactobacillus,bifidobacterium,enterococcus,bacteroides,escherichia coli.Results: At the tenth weekend,the serum TC,TG,LDL-C,ALT,LPS,IL-6, TNF-α ofgroup B and group C increased than initial (P <0.01).The number of fecallactobacillus,bifidobacterium,enterococcus,bacteroides of the2groups decreased thaninitial (P <0.01),while the number of fecal escherichia coli increased(P<0.01).The serumTC,ALT,LPS,IL-6of group D had no compared with initial(P>0.05),while TGdecreased(P<0.01). LDL-C,TNF-α increased than initial,but were still lower than that ingroup B and group C(P<0.05).The number of fecal escherichia coli of group D decreasedcompared with initial.The number of fecal lactobacillus,bifidobacterium,enterococcusdecreased,but were still higher than that in group B and group C(P <0.05). At the sixteenthweekend,the serum TC,TG,LDL-C,ALT,LPS,IL-6,TNF-αof group C decreased than at thetenth weekend and were also lower than group B(P <0.01).The number of fecallactobacillus,bifidobacterium,enterococcus of group C were higher than groupB(P<0.01),and the number of the fecal lactobacillus increased compared to the the tenthweekend(P <0.01).Conclusion: Feeding with high-fat diet can increase the SD rats’serum lipids, therelease of inflammatory cytokine and the change of intestinal flora.Saccharomyces boulardii has a certain degree preventive effect on the formation of hyperlipidemia,and canstabilize the intestinal flora, reduce the release of inflammatory cytokine.To the rat withhyperlipidemia,Saccharomyces boulardii can reduce the levels of serum lipids,improveintestinal flora and reduce the levels of endotoxin and inflammatory cytokines.
