Objective1. To analyze the changes of expression of Histamine H2、H4Receptor(H2R, H4R) in peripheral blood mononuclear cells(PBMCs) between Children with Henoch-Sehonlein purpura(HSP) in acute phase and convalescence, healthy children, discuss the correlation among these indicators and investigate their clinical significance.2. To analyze the changes of expressions of Histamine H2, H4Receptor(H2R, H4R) in peripheral blood mononuclear cells(PBMCs) between Children with Henoch-Schonlein purpura nephritis(HSPN) and the simple Henoch-Schonlein purpura nephritis(NHSPN), discuss the correlation among the histamine receptor and Henoch-Schonlein purpura nephritis.3. To compare to the histamine H2receptor antagonist cimetidine, histamine H2R changes before and after the treatment, to analyze the perspective of clinical study of histamine H2receptor antagonist effects on Henoch-Schonlein purpura histamine receptor expressions.4. To compare serum levels of interleukin-8, between Children with Henoch-Sehonlein purpura (HSP) in acute phase and convalescence, Healthy children, discuss the correlation among these indicators and investigate their clinical significance.5. To analyze the serum levels of interleukin-8between Children with Henoch-Schonlein purpura nephritis(HSPN) and the simple Henoch-Schonlein purpura nephritis(NHSPN), discuss the correlation among these indicators and investigate their clinical significance. Methods1. The expression of Histamine H2, H4Receptor(H2R, H4R)To analyze the changes of the expression of Histamine H2, H4Receptor in peripheral blood mononuclear cells between Children with Henoch-Sehonlein purpura in acute phase and convalescence, Healthy children.32patients with Henoch-Sehonlein purpura,32Children with Henoch-Sehonlein purpura in convalescence, and32volunteers were recruited in our study. Real-time fluorescent quantification PCR was used to detect the protein and mRNA expression of H2R, H4R on peripheral blood mononuclear cells. 2. Children with Henoch-Sehonlein purpura to histamine H2receptor antagonist cimetidine histamine H2R expression before and after treatment.Choose to our hospital for children with acute Henoch-Sehonlein purpura is divided into cimetidine group and no cimetidine group, Real-time fluorescent quantification PCR were used to detect the protein and mRNA expression of H2R, H4R on peripheral blood mononuclear cells.3. The serum levels of interleukin-8Western blot was used to detect the serum levels of interleukin-8between Children with Henoch-Sehonlein purpura(HSP) in acute phase and convalescence, Healthy children, to analyze the relationship between the IL-8and Henoch-Sehonlein purpura.The data were analyzed by SPSS18.0software. Results1. The protein and mRNA expression of Histamine H2, H4Receptor(H2R, H4R) in peripheral blood mononuclear cells(PBMCs) of children with Henoch-Sehonlein purpura in acute phase is higher than convalescence children and Healthy children(P<0.05). The mRNA expression of H2R, H4R in the HSP acute group were significantly higher than that in the convalescence and healthy control group(P<0.05). While no significant differences of Children with Henoch-Sehonlein purpura in convalescence, healthy children(P>0.05). The changes of expression of Histamine H2, H4Receptor(H2R, H4R)in peripheral blood mononuclear cells(PBMCs), no significant differences of Children between Children with HSPN and NHSPN in acute phase(P>0.05).To children with Henoch-Sehonlein purpura according to whether have hematuria and proteinuria, divided into abnormal urine test of no henoch-schonlein purpura nephritis group (NHSPN group) and henoch-schonlein purpura nephritis (HSPN group). The H2R mRNA level in children with nephritis group is (1.69±0.52), the mRNA level of nephritis group is (1.88±0.47) and the H2R mRNA level of healthy controls group is(1.22±0.18). H4R mRNA level in children with non nephritis group is (4.59±1.34), nephritis group H4R mRNA level is (4.39±0.53) and healthy controls H4R mRNA level is (1.44±0.33). The nephritis, purpura nephritis group H2R expression levels were significantly higher than that of healthy controls (p<0.05), the nephritis group and nephritis group H2R expression level no obvious difference (p>0.05). No allergic purpura nephritis group, nephritis group H4R expression levels were significantly higher than that of healthy controls (p<0.05), no allergic purpura nephritis group and nephritis group H4R expression quantity had no significant difference between the two groups (p>0.05).2. Cimetidine group the H2R mRNA level before treatment(1.77±0.51), than cimetidine group the H2R mRNA level before treatment (1.82±0.50) and healthy controls the H2R mRNA level (1.22±0.18), cimetidine group and cimetidine H2R level before treatment were higher than healthy controls (p<0.01). But cimetidine group and cimetidine H2R mRNA expression before treatment with no significant difference (p>0.05), and can be reference significance. After treatment, cimetidine group after treatment of the H2R mRNA level(1.07±0.05), than cimetidine group after treatment the H2R mRNA level(1.44±0.34), the two groups after treatment of the H2R mRNA expression level had significant difference(p<0.05), the group treated with cimetidine H2R mRNA level change is bigger.3. The serum levels of IL-8of children with Henoch-Sehonlein purpura(HSP) in acute phase is higher than convalescence children and Healthy children(P<0.05). The serum levels of IL-8in HSP acute group were significantly higher than that in the convalescence and healthy control group(P<0.05). While no significant differences of Children with Henoch-Sehonlein purpura in convalescence, healthy children(P>0.05).The serum levels of IL-8in children with HSPN in acute phase were significantly higher than that in the Children with NHSPN(P<0.05). Conclusions1. Henoch-Sehonlein purpura in children with acute mononuclear cells of histamine H2R, H4R protein and mRNA expression were significantly higher than that of children with Henoch-Sehonlein purpura recovery and healthy children. Children with the treatment of symptoms of Henoch-Sehonlein purpura convalescence H2R, H4R expression can drop to the level of healthy children. Histamine H2R, H4R onset process may be involved in Henoch-Sehonlein purpura.2. The acute phase of Henoch-Sehonlein purpura nephritis, nephritis group mononuclear cells in children with histamine H2R, H4R protein and mRNA expression were higher than healthy children, not Henoch-Sehonlein purpura nephritis group and nephritis group H2R, H4R expression level between the two groups have no obvious difference. Whether the kidney, is affected by Henoch-Sehonlein purpura, no obvious difference was found between the expression of histamine H2R and H4R. 3. For children with acute Henoch-Sehonlein purpura in the H2receptor antagonist cimetidine treatment group before and after treatment of histamine H2R changes more than cimetidine treatment way histamine H2R changes have significant differences, cimetidine can reduce histamine receptor expression.4. Children with acute Henoch-Sehonlein purpura significantly higher than the level of IL-8children with Henoch-Sehonlein purpura convalescence, healthy children After the treatment of symptoms of allergic purpura convalescence IL-8children can drop to the level of healthy children. IL-8disease process may be involved in Henoch-Sehonlein purpura.5. Children with allergic purpura nephritis is higher than the level of serum IL-8nephritis group, IL-8may participate in the occurrence of purpura nephritis and development.
