The Combined Treatment Of BMSCs Transplantation And Ea Stimulation Effects The BMP、 Neuroglial Cells And Blood Vessels Regeneration In ICH Rats

Objective:To discuss the combination of bone marrow mesenchymal stem cells transplantation and electric acupuncture effecting the expression of bone morphogenetic protein-2(BMP2)、 neuroglia antigen-2(NG2)、galactocerebroside(GALC)、CD68and laminin(LN) in intracerebral hemorrhage rats. Methods:100ICH models of male SD rats were established and randomized to divide into groups for the combined treatment of BMSCs transplantation and electric acupuncture(BMSCs+Ea group)、BMSCs transplantation group(BMSCs group)、electric acupuncture group(Ea group) and normal saline group (NS group) with each group having25rats. Each group was given the corresponding treatment respectively three days later, and divided into five subgroups for1d、3d、5d、7d and14d according to feeding time, with each subgroup having5rats. Make the gotten brains to frozen section and detect the expression of BMP2, NG2, GALC, CD68and LN by immunohistochemistry. Results:1. Statistical analysis for the BMP2positive cells around hemorrhage:The BMP2positive cells of each group showed a progressive increase constantly until to the peak at the7th day and then declined. The BMP2positive cells of BMSCs+Ea group were more than the NS group at the1st day~14th day(P<0.05). The BMP2positive cells of the NS group were fewer than the BMSCs group at the3rd day~5th day (P<0.05).There were no significant differences between the Ea group and NS group. The BMP2positive cells of Ea group declined at the1st day、5th day and7th day compared with the BMSCs+Ea group, and reduced compared with the BMSCs group at the1st day and5th day (P<0.05). The BMP2positive cells of BMSCs+Ea group rised compared with the BMSCs group at the1st day and14th day (P<0.05).2. Statistical analysis for the NG2positive cells around the hemorrhage. The NG2positive cells of the BMSCs+Ea group rised at the1st day~7th day compared with the NS group(P<0.05). The NG2positive cells of the BMSCs group were more than the NS group at the5th day and14th day (P <0.05). There were no significant differences between the Ea group and NS group. The NG2positive cells of the BMSCs+Ea group increased the relative to the Ea group at the1st day,3rd day,5th day and14th day(P<0.05). The NG2positive cells of the BMSCs group rised at the5th day and14th day compared with Ea group (P<0.05). The NG2positive cells of BMSCs+Ea group were higher than the BMSCs group at the1st day~5th day (P<0.05).3. Statistical analysis for the GALC positive cells around the hemorrhage. Compared with the GALC positive cells of the NS group in, the BMSCs+Ea group rised at the1st day~7th day, the BMSCs group presented a rise at the1st day~7th day, and the Ea group were relatively higher at the7th day(P<0.05). Compared with the GALC positive cells of the Ea group, the BMSCs+Ea group increased markedly at the1st day5th day, the BMSCs group rised at the5th day (P<0.05). The GALC positive cells of BMSCs+Ea group were more than the BMSCs group at the1st day~3rd day (P<0.05).4. Statistical analysis for the optical density(OD) value of CD68positive cells around the hemorrhage. The treatment group showed a progressive increase until to the peak at the7th day and then declined. The control group presented a progressive increase trend from1st day to the14th day. Compared with the OD value of the NS group, the BMSCs+Ea group rised at the1st day-14th day, the BMSCs group went up at the1st day-5th day and declined at the14th day, the Ea group was lower at the14th day(P<0.05). Compared with the OD value of the Ea group, the BMSCs+Ea group enlarged at the1st day~7th day(P <0.05).There was no significant difference in the OD value between the BMSCs group and Ea group. The OD value of the BMSCs+Ea group was higher than the BMSCs group at the5th day~7th day (P<0.05).5. Statistical analysis for the blood vessels around the hemorrhage: Compared with the blood vessels of the NS group, the BMSCs+Ea group rised at the5th day~14th day, the BMSCs group ascended at the3rd day~7th day,and the Ea group raised at the3rd day (P<0.05).Compared with the blood vessels of the Ea group, the BMSCs+Ea group increased at the5th day~14th day, the BMSCs group rised at the5th day~14th day (P< 0.05). The blood vessels of BMSCs+Ea group were more than the BMSCs group at the7th day (P<0.05). Conclusions:1. The combined treatment of BMSCs transplantation and Ea stimulation can play up-regulation to the expression of BMP2protein compared with the simple BMSCs transplantation and Ea stimulation at the ICH rats early. This helped NSCs and BMSCs to differentiate nerve cells and brain tissue remodeling.2. The combined treatment which can promote the proliferation of the OPCs and OLs had advantageous to remyelination; The proliferation of the OPCs and OLs can be inhibited in the late. This function maybe associated with preventing glial scar formation.3.The treatment of BMSCs transplantation activated the microglia and promoted it proliferation quickly. The effect of the combined treatment increased significantly and participated in immune reaction actively.4.The treatment of BMSCs transplantation and Ea stimulation can promotes the regeneration of blood vessels. The combined treatment had advantageous to angiogenesis and played a important role in improving the microenvironment of the hemorrhage and the surrounding area.
