Part I Expression of β-catenin, cyclinDl, c-myc of the Wnt signaling pathway in ovarian serous carcinoma and its clinical significanceObjective To study the expression of β-catenin, cyclinD1and c-myc of the Wnt signaling pathway in ovarian serous carcinoma and analyze its relationship between them and FIGO stage, pathological grade and prognosis. Also finding out wether there is a correlation between the β-catenin and cyclinDl, the β-catenin and c-myc.Methods The expression of β-catenin, cyclinD1, c-myc were examined in60ovarian serous carcinoma with immunohistochemistry PV methods. Chi-square test was adopted to analyze the relationship between β-catenin, cyclinD1and FIGO stage, pathological grade.Results (1)The positive expression of β-catenin was correlated with FIGO stage of carcinoma and the expression in stage III-IV was greater than in stage Ⅰ-Ⅱ (P=0.003); no statistical significance was found with pathological grade(P=0.817); lower survival rate of the patients was found in the group of positive expression of β-catenin (P=0.034), but it had no relationship with prognosis in advanced carcinoma (stage Ⅲ-Ⅳ)(P=0.495).(2) The positive expression of cyclinD1had no relationship with FIGO stage (P=0.829), it was correlated with pathological grade and the expression in high grade was greater than in low grade(P=0.017), lower survival rate of the patients was found in the group of positive expression of cyclinD1(P=0.009), and the same in stage Ⅲ-Ⅳ(P=0.039).(3) The positive expression of c-myc had no relationship with FIGO stage (P=0.069), it was correlated with pathological grade and the expression in high grade was greater than in low grade(P=0.016), there is no relationship between c-myc and the survival rate of the patients(P=0.266).(4)The expression of β-catenin had a positive correlation with the expression of cyclinD1, c-myc(P=0.014,P=0.005).Conclusions β-catenin and cyclinD1of the Wnt signaling pathway may play an important role in the development of the ovarian serous carcinoma, which may predict the poor prognostic for the patients. Examining the expression of them may be useful for our clinical work. Part Ⅱ The expression of β-catenin in ovarian cancer cell line COC2treated with all-trans-retinoic acidObjective The activation of Wnt/β-catenin signaling pathway may lead to cancer include the ovarian epithelial carcinoma. The key step of Wnt/(3-catenin signaling pathway is the accumulation of β-catenin in cytoplasm and enter the nucleus to promote the transcription of the related genes. β-catenin is the key molecules of Wnt/p-catenin signaling pathway. Our research was to explore the influence of ATRA on the expression of β-catenin and the mechanism of ATRA anti-proliferation effect in human ovarian cancer cell line COC2.Methods Ovarian cancer cell line COC2was treater with different ATRA concentration ranged from ATRA1,5to10μmol/L. We used RT-PCR and Western blotting to examine the mRNA and protein of P-catenin in the epithelial ovarian cancer cell line COC2treated with different ATRA concentrations after abstracted the total RNA of COC2cells in each group.Results The expressions of β-catenin between mRNA and protein were decreased in COC2cell line treated by ATRA, and present with dose-dependent manner.Conclusions ATRA down-regulated the expression of β-catenin in COC2cell line, this may suggest that the anti-proliferation effect for ovarian cancer cell treated with ATRA may relate to activation β-catenin pathway.
