| Objective:To observe the expression of proliferating cell nuclear antigen (PCNA)andbrain natriuretic peptide(BNP)in Pulmonary Artery Hypertension rats due toMonocrotaline(MCT) and to investigate its significance.Methods:40male SD rats were randomly divided into normal control group (n=8)ã€Model group (n=8) and treatment group (10mg Atorvastatin group,20mg Atorvastatingroup,Diltiazem group,each group n=8).The rats in Model and treatment group weregiven Monocrotaline (50mg/kg) by a single subcutaneously injection.Atorvastatin[10mg/(kg/d)] was given orally for3weeks to the rats in10mg Atorvastatin group sincethe day when rats were injected with Monocrotaline.Atorvastatin [20mg/(kg/d)] wasgiven orally for3weeks to the rats in20mg Atorvastatin group since the day when ratswere injected with Monocrotaline. Diltiazem [25mg/(kg/d)] was given orally for3weeks to the rats in Diltiazem group since the day when rats were injected withMonocrotaline. Normal control rats were just injected with constant volume saline.Atthe end of study, mean pulmonary arterial pressure(mPAP) and right ventricular systolicpressure were detected by cannulation through the right jugular vein.ELISA was usedto detect the plasm a level of brain natriuretic peptide.Proliferating cell nuclear antigen(PCNA) of pulmonary arterial vascular smooth muscle cell was detected with immunohistochemistry method.Results:1.MPAP and RVHI in Model group were more than that in normal controlgroup and treatment group(P<0.01). Compared with normal control group,MPAP andRVHI were increased in treatment group(P<0.01).MPAP and RVHI in treatment groupswere not different(P>0.05).2.PCNA and BNP in Model group were more than that in normal control group andtreatment group(P<0.01).Compared with normal control group, PCNA and BNP wereincreased in treatment group(P<0.01).PCNA and BNP in treatment groups were notdifferent(P>0.05).3.The expression of PCNA and plasma BNP levels in model group were positivelycorrelated(r=0.986,P <0.01),plasma BNP levels were positively correlated withpulmonary hypertension(r=0.889,P <0.01).Conclusion:Atorvastatin improve mean pulmonary artery pressure,right ventricularhypertrophy and Pulmonary vascular remodeling,partly due to inhibiting the expressionof proliferating cell nuclear antigen (PCNA)and brain natriuretic peptide(BNP)inPulmonary Artery Hypertension. |
