With the improvement of people’s living standard, in the new century, people’s diet industry is undergoing great changes, the public for food consumption concept has changed from the past to eat into a new level, to eat well at the same time, the problem of food security, especially microbial safety problems are gradually exposed, compared along with the traditional chemical class of antimicrobial agents for the natural plant source of antimicrobial agents has gradually become a new direction.Belong to Toona genus Meliaceae,Toona Sinesis is broad-leaved tree species.It has several familiar names,such as red cedar,Chinese toon,toon. Toona Sinesis is native to China, and it is distributed in Aisa and the Pacific.It is mainly grown in the yellow river and the Yangtse river, besides AnHui Province, ShangDong Province and HeNan Province etc.Toona Sinesis is the medically edible thing of dual purpose since ancient times in China.It is called "Green Health Food" abroad.Toona sinensis root bark, bark, buds, leaves, fruits and seeds are used medicinally, wherein the bud and leaf has the unique flavor, edible effect. According to traditional Chinese medicine, Chinese toon taste bitter, warm in nature, there is the treatment of dysentery, enteritis, gastric ulcer and other effects. Toona sinensis contain flavonoids, polysaccharides, saponins, alkaloids, oil and other chemical components.The extraction of effective chemical constituents in plants include ultrasound, extraction, microwave method, enzyme method, this paper mainly studies the old leaf of Chinese toon flavonoids, polysaccharides and saponins extraction, component analysis and determination of the antibacterial effect and the old leaf of Chinese toon application prospects were discussed. The following is the basic content of this paper research results:1.Using ethanol as solvent, coarse extraction of flavonoids in old leaves of Toona sinensis, with rutin as standard curve of total flavonoids were determined, and the extracts were removed antibacterial effect determination of ethanol, results show that the old leaf of Chinese toon flavonoids crude extract had a good antibacterial effect. AB-8macroporous resin were eluted with10%,30%,50%,70%different concentrations of ethanol, washed A, B, C, D four components.2.Methods for chemical composition of old leaves of Toona sinensis polysaccharide was extracted by ultrasonic extraction, using single factor test to determine the optimum extraction conditions of each single factor respectively:ultrasonic time45min, ultrasonic power240W, temperature60℃, under this condition the extraction rate of polysaccharide was5.49%old leaves of Toona sinensis. Preparation of standard curve using Coomassie brilliant blue G250method, the protein content were determined, and removal of protein by polyamide. The crude polysaccharide were studied after deproteinization, the filling material for the DEAE-Sepharose CL-6B ion exchange column, respectively with the concentration of NaCl solution for0.1,0.2,0.5mol/L phase gradient elution, three fractions were obtained. Named TSP1,TSP2,TSP3.3.The old leaf of Chinese toon saponin by chloroform-sulfiiric acid reaction,three triterpenoid saponins analysis for qualitative analysis.With70%ethanol reflux extraction4times, by petroleum ether, n-butanol, dissolved with a small amount of methanol, dropping the ether, obtained preliminary purification of crude saponin. The crude saponin extraction press optimization of single factor test and response surface, get the old leaves of Toona sinensis saponin verification rate of1.31%.4.Using filter paper method and the Oxford cup method, by bacteria (aureus, Escherichia coli, Bacillus subtilis), fungi (Aspergillus niger, yeast) as indicator bacteria on flavonoids, polysaccharides and saponins were antibacterial effect determination, results showed that the flavonoids, separation of components in A with antibacterial effect good for aureus and Escherichia coli, bacterial inhibition diameter were14.1mm,10.3mm, component D of Bacillus coli and aureus with good antibacterial effect of antibacterial diameter, respectively9.1mm,12.0mm; polysaccharide fractions of TSP1polysaccharide concentration25mg/mL inhibitory effect on E.coli obviously, TSP2the concentration of polysaccharide100mg/mL on aureus antimicrobial effect obviously, Bacillus subtilis TSP3more obvious bacteriostatic effect of50mg/mL I in polysaccharide concentration; saponin extract at the concentration of1mg/mL, obviously on Escherichia coli and Bacillus subtilis antibacterial effect.
