To Detective The Theory Of Zhuanggu Zhitong Prescription Inhibits Postmenopausal Osteoporosis From The Regulation Of The Cytokine Network

Objective:To detective the theory of zhuanggu zhitong prescription inhibits postmenopausal osteoporosis from it regulates the cytokine network of postmenopausal osteoporosis.Methods:Using60three month old SD female rats to establish the model of postmenopausal osteoporosis, after the ovariectimized surgery, then divided the rats randomly into the zhuang guzhitong prescription high concentration group(A), the media concentration group(B), the low concentration group(C),the model group(D),the estradiol valerate positive control group(E), the sham operation group(F)(n=12for each). Then intragastric administration after one week later, the intragastric administration of drugs of zhuanggu zhitong prescription high media low concentration groups are13.2mg/kg6.6mg/kg3.3mg/kg, the estradiol valerate positive control group is0.021mg/kg of estradiol valerate, and the sham operation group and the model group are intragastric administration distilled water for1ml/100g, ntragastric administration for13weeks. Then every group randomly chose6female rats to extract the mRNA sample of femur tissue and start to get PT-PCR test the expression of mRNA of IL-10IL-17TNF alpha TGF beta estrogen receptor beta and estrogen-related receptor alpha and6female rats to collect blood from abdominal aorta and centrifuge(500g) after solid and get the serum to use Elisa to test the concentration of IL-10IL-17TNF alpha TGF beta IFN gama and estradiol.Results:RT-PCR tests the expression of mRNA of femur tissue, compared with the model group, the expressiong of IL-10mRNA of all group had significantly increase; excepted the low concentration group, the expression of IL-17/TNF alpha/TGF beta/ER beta/ERR alpha of each goup had significantly increase. The enzymelinked immunisorbent assay test the concentration of cytokine and estradiol of serum. Compared with the model group, excepted the low concentration group and the estradiol valerate positive control group, the concentration of IL-10of had siginificantly increase, excepted the low concentration group, the concentration of IL-17/TNF alpha/estradiol of each group had significantly increase, the concentration of TGF beta and IFN gama of all group had significantly increaseConclusion:.1.zhuanggu zhitong prescription inhibits postmenopausal osteoporosis by the way of estrogen;2.zhuanggu zhitong prescription inhibits postmenopausal osteoporosis by the way of increase Estrogen receptor bata to increase the susceptibility of estragen;3zhuanggu zhitong prescription inhibits postmenopausal osteoporosis by the way of quasi hormone like effect to activate transmission of the Estrogen receptor bata signal;4.zhuanggu zhitong prescription inhibits postmenopausal osteoporosis by the dynamic equilibrium of bone resorption and bone formation of cytokine effection.
