Objective: To investigate cytotoxicity of poly(lactic acid) electrospun membranesloaded with cisplatin and chloroquine for oral squamous cell carcinoma CAL-27cellline.Methods: Prepared these membranes (DDP/PLA membranes、CQ/DDP/CQ/PLAmembranes and CQ/DDP/PLA membranes) by electrospinning. Then the micromorphology of these membranes were observed by scanning electronmicroscope(SEM); the degradation rate of PLA membrane was measured by UVspectrophotometric; the proliferation activity of CAL-27cells was assayed bytetrazolium bromide (MTT) colorimetry;At last,the LC3-Ⅱexpression level wasdetected by laser scanning confocal microscope.Results:1. Nanofibers was continuous、smooth and uniform diameter;2. BSA couldbe diffused out from the membranes slowly and uniformly. The membranes degratedtime was about21days;3. The MTT result showed: compared with the control group,at first, the effect of cell killing treat with DDP/PLA membranes group、CQ/DDP/CQ/PLA membranes group and CQ/DDP/PLA membranes group were notobvious; As the extension of time, the effect of cell killing treat with DDP/PLAmembranes group、 CQ/DDP/CQ/PLA membranes group and CQ/DDP/PLAmembranes group were enhanced(P<0.05).4. Immunofluorescence results showedthat the fluorescence intensity of CQ/DDP/CQ/PLA membranes group andCQ/DDP/PLA membranes group were lower than DDP/PLA membranes group.Conclusion:1.Obtain poly(lactic acid) electrospun membranes loaded withcisplatin and chloroquine that had sustained-release effect.2. DDP/PLA membranes、CQ/DDP/CQ/PLA membranes and CQ/DDP/PLA membranes had sustained-releaseeffect、maintained longer period of drug concentration and prolonged action ofchemotherapeutic agents.3. CQ/DDP/CQ/PLA membranes and CQ/DDP/PLAmembranes could down-regulated the level of autophagy and enhance killing effect against oral cancer CAL-27cells. |
