Pancreatic ductal Adenocarcinoma is a substantial health problem worldwide at present. Early diagnosis and judgement of prognosis could enhance quality of life and patient survival. The aim of this investigation was to identify the correlation with the expression(at level of genes and protein) of laminin-332(LM-332, formerly termed laminin-5) and clinicpathologic significance and determine their influence on survival in pancreatic ductal adenocarcinoma.The expression of LM-332α3/β3/γ2subunits mRNA in37groups pancreatic carcinoma specimens and non-tumor pancreatic tissues was investigated by QRT-PCR analyses. Using immunohistochemistry methods, the differential expression of the protein expression levels of three chains, respectively, was analyzed between96pancreatic carcinoma specimens and90non-tumor pancreatic tissues. The expression levels of proteins immunohistochemically in pancreatic ductal adenocarcinoma were investigated for associations with clinicopathological characteristics taken from patient data. Using the Kaplan-Meier method and compared by log rank analysis, Survival curves were performed.QRT-PCR result indicated that the Overall relative value by2-A(ACt) of LNa3mRNA and LNy2mRNA were increased compared with non-tumor tissues, but some of them were lost or down-regulated, which were not associated statistically(P=0.089and P=0.054respectively). Immunostained expression for LNa3and LNy2was found in all tumor tissues from96patients. The expression level of LNa3, LNβ3and LNγ2was associated with each other. LNa3、LNβ3and LNy2positivity were all significantly associated with differentiation and advanced stage (P<0.05). According to the location of LNγ2immunohistochemical expression patterns, samples were classified into three groups as follows:basement membrane type (B type); cytoplasmic type (C type); mixed type(M.type).B type was significantly corraleted with differentiation (well differentiated type)(P=0.010) and M type was significantly associated with hepatic metastasis (P=0.031). We concluded that patient outcomes for those with high expression of LNa3and LNy2were significantly worsen than for those with low expression(P=0.008; P=0.000); patient outcomes for those with all high expression of three subunits were significantly worsen than for those with other expression (P=0.000); pateints with B type of LNy2showed better outcomes campared to patients with the C type or M type (P=0.012;P=0.003)Our results suggest that the overexpression of α3/β3/γ2chains of laminin-332may be play a substantial role in the progression and prognosis of in pancreatic ductal adenocarcinoma.
