With the development of society and the improvement of science and technology, medical and health care is also steadily forward. Regarding "take medicine", time is different now, the public awareness has changed. It prompts the existing treatment being constantly perfected that people pay more attention to the rationality and validity of clinical medicine treatment. Meanwhile, with the national strong support of developing and producting drugs, the variety and quantity of medicine which is being used in clinical treatment now are increasing. Additionally, more reasons have led to the universality of drug combination therapy, for example, the more and more deep understanding of disease, the detailed classification of disease treatment or the changing attitude of doctors from the single treatment to the combination therapy. Although drug combination demonstrate the efficacy and reduce side effects in its positive side, there is no denying that it is also showing some adverse reactions and a certain degree of inapplicability of clinical use.Drug-drug interactions among the clinical commonly used drugs are universal, through literature research, the paper designs a research plan on metabolism-based drug-drug interactions focused on the inhibition of CYP450 enzymes. In addition, from the perspective of clinical drug combination, dexamethasone and diazepam have been selected as the case study objects, the HPLC-MS/MS methods have been established to determine the concentration of dexamethasone, diazepam and their metabolites in vitro, drug-drug interactions between dexamethasone and diazepam have been studied preliminarily, which lays the foundation for further study between dexamethasone and diazepam, even other drugs. Meanwhile, the case study verified the feasibility of the research plan, in order to make more drug studys and provide a reference to guide rational clinical drug combination.
