The Research Of Neutrophil Elastase Expressing In Rabbits ACST

Obstructive:Research NE expression and it’s significance in rabbits with acute severe cholangitis (ACST), explore NE and NEI role and mechanism of ACST.Methods:with 36 rabbits divided into the control group (sham group), the observation group (ACST group), the treatment group (ACST+NEI group) comparative study, respectively.at time of the experimental began 0h,3h,6h,12h,24h, 48h collecting blood and bile samples to Elisa measured the serum NE, IL-6 and bile NE content by UV rate method serum ALT levels, at 48h get liver, bile duct tissues of each group’s rabbit to pathological examination.Each group of measured data us statistical software SPPS17.0 make analysis and comparison.result:1, each time the control group have no significant difference in all the measured values, the observation group and the control group of rabbits measuring serum NE, bile NE,serum IL-6, ALT values increasing with time during the observation of 48h, NE, IL-6 increased the fastest in the first 12h, ALT rose fastest in 24h-48h. Measured values contrast with the control group P<0.01, have significant difference; 2, using NE inhibitor Sivelestat Carolina (treatment group) reduced expression NE in rabbits ACST, the serum IL-6, ALT also decreased accordingly the treatment group and the observation group contrast detection value P<0.05, have significant difference;3, each group of pathological observation showed that:In the observation group liver periportal and lobular has acute neutrophil infiltration, hepatocytes has focal necrosis, fatty degeneration, eosinophilic change.the bile duct has purulent inflammation, bile duct epithelial cells extensive necrosis and shedding. The observation group pathology was more serious than the treatment group and The control group no abnormalities.Conclusions:1, NE expression increased in the ACST, NE may provide diagnostic value to ACST, also reflect inflammatory status and severity.2, Use NEI sivelestat can effectively reduce the expression levels of NE in ACST, inhibit inflammation, protect the liver tissue. Proofed NEI have it’s effective treatment function to ACST.
