Objective:Research the differences between epirubicin micro pump into the abdominal cavity and intraperitoneal injection efficacy in the treatment of breast cancer in mice.Methods:1. Cultured mouse breast cancer cell line 4T1-luc, passaged the cell, using the SRB assay calculates Epirubicin(EPI) on 4T1-luc cells in vitro inhibition.2. Feeding Balb/C female mice, inoculation 4T1-luc cell suspension to breast pad order to construct mouse breast cancer model. Measuring the longest and shortest diameter of the mouse tumor using vermier caliper, then calculates tumor volume. Dividing the tumor mice into 3 groups when tumor diameter is 3mm.3. The same dose of epirubicin (8mg/kg) was intraperitoneally administered in different ways:: ①According to 8mg/kg table Epirubicin(EPI) use sterilization injiection water dissolve into the Osmotic pump and put it into rats abdominal cavity for experimental capsule (MP); ②intraperitoneal injection directly (El) for control group;③Blank group not be table Epirubicin(EPI) intervention, but injiecting sterilizing water 1ml into abdominal cavity.4. At the 6th day,12th day of the 4 t1- luc cell suspension inoculation on mouse pads giving EPI intervention; at 18th day giving fluorescence scanning imaging,then analysis the image and calculation the fluorescent photons.5. After the mice giving fluorescence scanning imaging, executed mice by neck method, separated transplantation tumor and heart, fixed in 10% formalin liquid, fixed after making the pathological slice, observated pathology differences after HE staining.Results:1. The volume of transplanted tumors in mice comparison results:two different ways of EPI delivery compared with the blank group showing certain tumor suppression effect, difference have statistical significance (P=0.000); Abdominal trace continuous dosing pump group with intraperitoneal injection of direct tumor suppression effect, experimental tumor volume is small, but the repeated measurement single factor analysis of variance has no statistical significance (P= 0.165).2. The mice in vivo fluorescence scanning imaging photon number comparison results:the experimental group (celiac trace continuous dosing pump group) and control group (direct intraperitoneal injection group), blank group bug luciferase injection after 5 min,10 min,15 min fluorescence scanning imaging, to compare two different time each group, the experimental group and the blank group differences were statistically significant (P5min=0.003, P10min=0.003, P15min=0.003); control group compared with the blank group differences were statistically significant (P5min=0.011, P10min=0.011, P15min=0.011);There are differences between the experimental group compared with control group, but no statistical significance (P5min=0.473, Piomin=0.473, P15min=0.206).3. transplantation tumor and heart pathologic results comparison:(ecause of pathological production failure, without pathology comparison results.Conclusions:Epirubicin peritoneal micro-pump group has the same efficacy with direct injection group.
