At Present, People had done a lot of researches in the development of cerebellar cortex, cell morphology, but during the developing Process of cerebellum cortical, on account of lacking of Reelin Protein effect on vascular development research and Produce of cell Polarity changes do not very clear. Reelin stop signal, as a kind of cell migration, about the formation and development, layers of cerebral cortex nerve cells migration,vascular development and cell Polarity Play a very important role. We use wild type(WT) and Reeler in mice with Nissl staining, vascular Perfusion and immunocytochemistry for experimental research, this is we are going to delve into the lack of Reelin Protein effect on cerebellar cortex and vascular development.Objective: Our aim is to observe the developing Process of mice cerebellar cortical and blood vessels as well as changes of nerve cell Polarity; to explore the relationship among the occurrence of blood vessels and cells in Reeler mice, analyse the change of Purkinje cell Polarity and the vascular development of wild type(WT) and Reeler in mice.Methods: The Nissl staining, vascular Perfusion and immunocytochemistry were used to take on morphological observation of the nerve cells development from embryonic(E17) to Postnatal mice(P21), compare and measure the size of the diameter and angle in Purkinje Positive cell number of the Purkinje cell layer and cerebellar cortex vascular volume density in wild type(WT) and Reeler in mice.Results: â‘ In the Process of cerebellar cortical layers development in Reeler mice, cerebellar cortical lamella do not form a normal three layer: external granular layer(molecular layer), Purkinje cell layer, internal granular layer. â‘¡ Purkinje cells Polarity has changed during the migration Process, compare and measure the size of the diameter and angle in Purkinje Positive cell number of the Purkinje cell layer, there has much significant differences of WT and Reeler mice cerebellum in Purkinje cell Polarity, P < 0.05. â‘¢ Ink Perfusion method showed that vascular volume density in the cerebellum was on a rising trend, correlation analysis indicated that at P3, P7,(P < 0.05), P14, P21,(P < 0.01), and it showed that age of mice and vascular density is highly relevant, there has significant difference statistically.In addition, the study also showed that there has significant difference in vascular morphology in wild type(WT) and Reeler in mice.Conclusion: In the Process of cerebellar development, because of lacking of Reelin Protein, cerebellar cortical lamella do not form a normal three layer, nerve cell Polarity is changed, the arrangement of blood vessels are disorders, compared with the same age of wild mice, the volumetric density of vasculature in the cerebellum of Reeler mice increase. In the Process of cortex evolution, Reelin Protein Play an important role, Reelin control the cortical neurons morphology change and migration direction; Reelin maintain the cortex of mammals development from inside to outside; Reelin formate the normal three-layer structure; Reelin, as scaffold Protein, adjust the Parallel arrangement of the cortex radial glial cells, Provide support of correct neurons migration and maintain the normal blood vessel development. Reelin Protein mutations can lead serious damage of the development of the cortex,cortical layers developmental disorders, blood vessel disorders as well as the nerve cell Polarity change. |
