Objective:To investigate in vitro percutaneous characteristics of compound lidocaine cream,and the consistenty of in-vitropercutaneous with the Emla cream.Todesignand optimize prescription, and determine the amount of each component in the prescriptionforhomemade compound lidocaine cream in laboratory.Methods:Lidocaine and prilocaine were quantified by HPLC;The in vitro percutaneous penetration testexperiments of compound lidocaine cream through hairless mouse skin,big abdomen skin and bama miniature pigs were perfromed by modified Franz’s cell. To investigatecompound lidocaine cream rheology using the properties of multifunction texture analyser,and to screen and optimize prescription by percutaneous penetration testin vitro.Results:There were good linear relationship between the peak area and concentration in the range of a certain concentrationfor lidocaine and prilocaine. The recovery and precision could meet the requirements. Percutaneous characteristicsof lidocaine and prilocaine accord with zero order kinetics equation.the transdermal rate for reagent and controlled drugs lidocaine is respectively34.53% and 2.37%in 8 h by rat,the transdermal rate for reagent and controlled drugs prilocaine is respectively39.55% and 37.71%in 8 h by rat.the transdermal rate for reagent and controlled drugs lidocaine is respectively(0.113±0.0080)% and(0.107±0.0064)% in 1 h by pigskin,Retention rate is respectively( 1.219±0.0579) % and( 1.196±0.0552)%.the transdermal rate for prilocaine is respectively(0.096±0.0076)%and(0.0915±0.0055)% in 1 h by pigskin,Retention rate is respectively(0.982±0.0688)%and(1.003±0.063)%.the transdermal rate for reagent and controlled drugs lidocaine is respectively(0.519±0.0375)%and(0.514±0.0218)% in 2 h by pigskin,Retention rate is respectively(1.794±0.0876)% and(1.745±0.0479)%.the transdermal rate for prilocaine is respectively(0.439±0.0409)%and(0.427±0.0149)%in2 h by pigskin,Retention rate is respectively(1.564±0.140)%and(1.530±0.0892)%.There were no significant difference(p > 0.05) in percutaneous rate and retention rate between test and control creams by t-test statistics. the amount of carbomer in the homemadecompound lidocaine cream prescription is 1.2%,the amount of polyoxyethylene hydrogenated castor oil is 0.5%,the amount of Sodium hydroxide is 0.5%.Conclusion:In vitropercutaneous patternare consistent for test and control compound lidocaine creamsthrough big abdomen skin and bama miniature pigs. The skincumulative percutaneous quantity and rheological characteristics are consistent for homemade and control compound licocaine creams.
