Objective:Explore matrine injection by TGF-β1-induced human embryonic lung fibroblast proliferation, apoptosis and cytokine expression, to explore the therapeutic effect on pulmonary fibrosis and the mechanism study.Methods: Human embryonic lung fibroblast cells were divided into five groups, a control group, TGF-β1(1ng / ml) in the model group, TGF-β1(1 ng / ml) + matrine injection(10,20,40 ul / mL) for intervention therapy. Application of MMT assay in human embryonic lung fibroblast proliferation; flow cytometry AnnexinV / PI double staining apoptosis rate; semi-quantitative RT-PCR was used to detect COL- ⅠmRNA, COL- ⅢmRNA, α-SMA mRNA, ECSOD mRNA, PDGF mRNA; ELISA assay culture supernatants COL-Ⅰ, COL-Ⅲ, α-SMA, ECSOD, PDGF content.Results: 1.MTT Detection: model group, cell proliferation the absorbance(OD) value was significantly higher than the control group, reducing cell proliferation Matrine absorbance(OD) value compared with the model group, and with greater magnitude lower concentration increased, there was a significant differences(P <0.05); 2. Flow cytometry Annexin V / PI double staining: the model group compared with the control group, the apoptosis rate was significantly lower, Matrine apoptosis compared with the model group was significantly increased, and the drug concentration the higher, the higher the rate of apoptosis, there was a significant difference(P <0.05); 3.RT-PCT test: the model group compared with the control group COL- mRNA, COL-Ⅲ ⅠmRNA, α-SMA mRNA, PDGF mRNA expression was significantly increased, mRNA expression ECSOD lower, compared with the model group, matrine injection group COL- mRNA, COL- mRⅢ ⅠNA, α-SMA mRNA, PDGF mRNA expression was significantly reduced, ECSOD m RNA expression increased, and the drug the higher the concentration, the lower / higher the larger the magnitude, there was a significant difference(P <0.05); 4.ELISA assay: the model group compared with the control group, cell culture supernatants HELF COL-Ⅲ, COL- Ⅰ, α-SMA, PDGF levels increased, ECSOD reduce matrine injection group compared with the model group COL-Ⅲ, COL-Ⅰ, α-SMA, PDGF were significantly reduced, ECSOD significantly higher, and the higher concentration of the drug, which reduces / increase in magnitude greater, there was a significant difference(P <0.05).Conclusion:Matrine injection induced by TGF helf cells, inhibit proliferation and promote apoptosis, inhibition of lung fibroblast collagen synthesis, inhibit fibroblast to muscle into fiber cell transformation, anti-oxidation, inhibition of platelet derived growth factor, and a certain range of concentration higher stronger role.
